In the United States, the first ironworks was built at Jamestown, Virginia, in 1619. The Hammersmith furnace in Saugus, Massachusetts, built in 1645, operated until 1675. This early American ironworks has been restored and is called the Saugus Iron Works. Iron blast furnaces appeared in many locahties where there were deposits of iron ore. The agar [9002-18-0] plate method consists of adding a known quantity of sample, usually 1.0 or 0.1 mL, depending on the concentration of bacteria, to a sterile petti plate and then mixing the sample with a sterile nutrient medium. Pewter. Modem pewter may have a composition of 90—95 wt % tin, 1—8 wt % antimony, and 0.5—3 wt % copper. Lead should be avoided by contemporary craftsman because it causes the metal surface to blacken with age. Pewter metal can be compressed, bent, spun, and formed into any shape, as weh as being easily cast. A wide variety of consumer articles ate available from domestic and foreign manufacturers. Reproductions of pewter objects from colonial times, some cast from the original molds, ate popular. The annual U.S. production of pewter exceeds 1100 t. [Pg.62]

One standard test used to determine the presence of the coliform group is called the multiple-tube fermentation technique (sometimes called the presumptive test). If this test indicates the presence of these bacteria, then a confirmed test must be done. If only negative colonies or no colonies develop during this test, it is considered negative otherwise, a completed test must be undertaken. Positive results obtained in the completed test are evidence for the presence of coliform bacteria. Testing methods have been given by the APHA, and the detailed procedures contained therein should be consulted (20). [Pg.233]

Another standard test, which is much simpler and more convenient, is the membrane filter technique. A suitable volume of sample is filtered through a sterile, 0.45-p.m membrane filter. The filter is placed in a petri dish containing a specific growth medium (M-Endo nutrient broth, M-Endo medium) and incubated for 24 h at 35°C. If after this time the colonies show the characteristic green sheen, this is taken as positive evidence for the presence of the coliform group (see Water, sewage). [Pg.233]

Low and high refer to the number of colony forming units (CPU) used to inoculate the media. Comparing MICs at low (10 ) and high (10 ) CPU, a large increase in MIC at the high inoculum level vs a P-lactamase producer is an indicator of instability via enzymatic hydrolysis. [Pg.65]
