Microbial colonies

Just as there are many different types of systems, there are many different types of transforms. In the algebraic system pictured in Figure 1.2, the system transform is the algebraic relationship y = a + 2. In the wine-making system shown in Figure 1.3, the transform is the microbial colony that converts raw materials into a Hnished wine. Transforms in the chemical process industry are usually sets of chemical reactions that transform raw materials into finished products. [Pg.14]

Count the number of microbial colonies on each plate. The microbial estimates are reported per contact plate or per swab. Examine the types of colonies and submit the representative colonies for identification. Mold does not carry special status relative to bacteria. Any significant shifts in type or number require action — regardless of mold vs. bacteria. [Pg.767]

It should be mentioned that most natural aldolase enzymes can also be assayed using enzyme-coupled systems relaying the reaction to a redox process with NAD. The formation of NADH by active microbial colonies in expression libraries of mutant enzymes was detected colorimetrically in agar plates using phenazine methosulfate and nitroblue tetrazolium, which forms an insoluble precipitate. The assay was used by Williams et al. [14] and Woodhall et al. [15] for evolving sialic acid aldolases to accept non-natural aldehyde acceptors. [Pg.4]

Several studies have combined results from different MSI approaches or to integrate them with other, non-MS-based imaging techniques. Tellurium may also be transformed by reduction and methylation (Chasteen and Bentley, 2003). The ASTM procedure determines an ability of microbes to stick to the surface of polymeric materials (containing incorporated biocides) and continue to inhabit it. The same procedure can be applicable to composite materials. Clearly, if the composite contains biocide, which diffuses to the surface, it would diminish microbial colonies on the surface. If, on the contrary, biocide is firmly immobilized within the matrix (e.g., if it is practically water-insoluble and does not move to the surface), it would not prevent microbial staining. Obviously, a biocide should not be too water-soluble and promptly leached out of the material, and it should not be practically irreversibly bound within the matrix. Actually, if the biocide quickly leaves the matrix, such as wet deck boards, the ASTM procedure would also be helpful in determining this fact. [Pg.439]

ASTM E 2180 aims at comparing counts of microbial colonies at the surface in a control (untreated) sample and a sample made in the presence of biocides and incorporated into the matrix. A sample can be either freshly produced, or washed with water over a certain time period, or taken from the field. [Pg.439]

Soils vary spatially in Eh values, and thus a large number of measurements should be taken to adequately characterize a given area. This variability could be minimal in permanently waterlogged wetlands or areas with minimal hydrologic fluctuations. A platinum electrode inserted into the soil may be in direct contact with the organic matter or other solid phase, which may result in lower Eh values than those for the soils as a whole. The fractal dimension of the surface Ds (boundary/interface) of a solid structure is obviously an important characteristic, but many of the physical properties of solids also depend on the scaling behavior of the entire solid and/or of its pores.
