Broth nutrient

Another standard test, which is much simpler and more convenient, is the membrane filter technique. A suitable volume of sample is filtered through a sterile, 0.45-p.m membrane filter. The filter is placed in a petri dish containing a specific growth medium (M-Endo nutrient broth, M-Endo medium) and incubated for 24 h at 35°C. If after this time the colonies show the characteristic green sheen, this is taken as positive evidence for the presence of the coliform group (see Water, sewage).

To 100 ml of a sterile nutrient broth (composed of Bacto-beef extract, 3 g Bacto-peptone, 5 g per liter of tap water) in a 300 ml flask is added one loopful of the incubated culture and the broth mixture is further incubated for 24 hours at 28°C on a shaking machine. 

The broth culture so obtained is employed as an inoculum (1%). Into each of ten flasks containing 100 ml of sterile nutrient broth is added 1 ml of the inoculum. The flasks are agitated on a rotary shaker for 8 hours at 28°C at 240 strokes per minute. After this growth period, a solution of 25 mg of 16/3-methy(cortisone in 0.5 ml of methanol is aseptically added to each flask which in turn is reshaken and incubated for an additional 24 hours. The final pH is 7.8. 

In each cycle, the library of mutated genes is first inserted in a standard bacterial host such as Escherichia coli or Bacillus subtilis. Subsequently, bacterial colonies are plated out on agar plates and harvested individually by a colony picker. Each colony is placed in a separate well of a microtiter plate containing nutrient broth, so that the bacteria grow and produce the protein of interest. Because each colony originates...

Chthoniobacter flavus has been obtained in pure culture (Sangwan et al. 2004), which was made possible by the use of a dilute nutrient broth medium solidified with gellan gum in place of agar (Janssen et al. 2002), and this has since been extended (Sangwan et al. 2005).

Appropriate Xenorhabdus or Photorhabdus bacterial cells are aseptically transferred to 5 ml of nutrient broth in a test tube and kept overnight on a shaker. The flasks containing autoclaved material are inoculated with the bacterial culture by pouring the contents of one culture tube. The flask is shaken well and stored for 2-3 days at 25°C to allow multiplication of the bacteria.

Each selected test strain is grown for 10 h at 37°C in nutrient broth (Oxoid No. 2) or supplemented minimal media (Vogel-Bonner) on an orbital shaker. A timing device can be used to ensure that cultures are ready at the beginning of the working day.

Certain bacteria are selected based on their ability to attach to specific metals. These bacteria are then grown in a nutrient broth and then placed in contact with a porous synthetic fiber such as Dacron , Orion , or Tyvek fiber. Wastewater is then brought into contact with these bacteria-coated fibers, and specific metals in the wastewater attached to the bacteria on the fibers. The metals are removed from the fibers through washing, burning, contact with sodium carbonate, or by some means.
