Colony assay

Poly(DL-lactide) was used as the excipient in microspheres of CCNU, a nitrosourea, prepared by a solvent evaporation procedure (96,97). PLA-CCNU microspheres 3.0 pm in diameter were injected i.v. and leukemia cell survival was determined by spleen colony assay. A 100-fold decrease in leukemia cell survival was observed with the microspheres in both spleen and liver compared to untreated controls. Promising results were also obtained with Lewis lung carcinoma in mice. These studies showed that 2- to 4-ym microspheres were preferentially targeted to the lungs.

The conditioning of the CD34+ cells as analyzed by colony assays in methylcellulose revealed that membrane-bound SCF mainly supports the maintenance of multipotent and bipotent progenitors. The number of CFU-GM and CFU-GEMM constituted 32% of total colony-forming cells after one week and 52% after two weeks in coculture. In contrast, the proportion of multipotent progenitors in cocultures induced by SI SCF are 4-fold lower after three weeks in culture (Table 2), but the contents of erythroid colonies (BFU-e s) increased 1.5-fold at the same time point. The results indicate that soluble SI SCF promotes more committed cells than membrane-bound SCF. Mature myeloid colonies increased about 1.5-fold in contrast to CFU-C derived from membrane-bound SCF expressing cultures (Friel et al, 2002).

A number of cytokines (see p. 280) stimulate the growth, differentiation and functional activity of myeloid progenitor cells. As the name implies the function of these polypeptides was defined by in-vitro colony assays of bone marrow progenitors. They have effects on all myeloid cells including the multipotential stem cells (but probably not the more immature pluripotentiai cells), intermediate progenitors and circulating mature cells. Those in clinical use are described below.

Since the colony assay cannot be used to measure stan and progenitor cell proliferation parameters, a different readout was required.
