Ibuprofen 42

Ibuprofen is an over-the-counter drug that is widely used as an antiinflammatory and a general analgesic, where it competes with aspirin. [Pg.67]

Worldwide annual production is of the order of 7000-8000 tons which is very large for a pharmaceutical. [Pg.67]

There are basically two processes for the production of ibuprofen (see Fig. 2.31). Both involve a Friedel-Crafts acylation as the first step. In the classical route, developed by Boots, the p-isobutylacetophenone is subsequently converted to ibuprofen in five steps involving conventional organic chemistry. In the catalytic route, more recently developed by Hoechst, the p-isobutylacetophenone is converted in two steps. [Pg.67]

The first step is catalytic hydrogenation to the corresponding alcohol over a 5% palladium-on-charcoal catalyst in methanol solvent at 30°C and 7 bar. The selectivity is 97% at 99% conversion. In the second step the alcohol is carbonylated at 35 bar CO and 125-130°C in the presence of a PdCl2(Ph3P)2 and HC1 as catalyst. Selectivities to ibuprofen are ca. 70% at 99% conversion. [Pg.67]

The catalytic route has the advantages of being shorter and producing considerably lower amounts of inorganic salts than the classical route. [Pg.67]

Ibuprofen is a common pain reliever that (even in low dosages) used to cause a false THC positive on the EMIT test. The EMIT has been changed to use a different enzyme to eliminate false positives due to Ibuprofen. Ibuprofen in very high doses will still interfere with both the EMIT and the GC/MS. There is some conflicting data here because some sources say that the GC/MS tests can distinguish between Ibuprofen and THC (as well as other over-the-counter drugs). [Pg.31]

Like aspirin, ibuprofen is a nonsteroidal anti-inflammatory drug. It is a cyclooxygenase inhibitor that interferes with COX-1 and COX-2 forms of that enzyme. Its effects on COX-2 give it fever-reducing (antipyretic), analgesic (pain relief), and anti-inflammatory functions. [Pg.183]

Its effects on COX-1 give it several undesirable effects it is an anticoagulant, and it irritates the stomach lining. [Pg.184]

Ibuprofen is used to relieve fever and pain, and symptoms of arthritis and menstrual cramping. It may also reduce the dizziness associated with standing up quickly (called orthostatic hypotension), and some studies have shown that it may have beneficial effects for patients suffering from Alzheimer s disease and Parkinson s disease. [Pg.184]

Devise and perform an extraotion procedure fhat may be used to separate pure 2-(4 -isobutylphenyl)propionic acid (ibuprofen) from the over-the-counter medications Advil or Nuprin . Determine the melting point of the material you isolate to oonfirm its identity. [Pg.167]

Place the used filter papers in a container for nontoxic solid waste. Flush the acidic and basic filtrates down the drain. Pour any diethyl ether that has been isolated into a container for nonhalogenated organic liquids. Pour any dichloromethane that has been isolated into a container for halogenated organic liquids. [Pg.167]

Using Equation 5.9, show that three extractions with 5-mL portions of a solvent give better recovery than a single extraction with 15 mL of solvent when K = 0.5. [Pg.167]

Considerable advances in asymmetric hydroformylation, a process which, among other things, provides a potential route to enantiomericaHy pure biologically active compounds, have occurred. Of particular interest are preparations of nonsteroidal antiinflammatory (NSAI) pharmaceuticals such as Naproxen (8) and Ibuprofen (9), where the represents a chiral center. [Pg.471]

From 1981 to 1995 other dmgs switched to OTC were the antifiingal preparations containing micona2ole nitrate [22832-87-7] (7) and clotrima2ole [23593-75-1] (8) the antiinflammatory agent ibuprofen [15687-27-1] (9) and the histamine H2-receptor antagonists famotidine [76824-35-6] (10) and cimetidine [51481-61-9] (11). [Pg.226]

Nonsteroidal Antiinflammatory Drugs. Nonsteroidal antiinflammatory dmgs (NSAIDs) include, among the numerous agents of this class, aspirin (acetylsaflcyhc acid), the arylacetic acids indomethacin and sulindac, and the arylpropionic acids, (5)-(147) and (R)-(148) ibuprofen, (5)-(149) and (R)- (150), flurbiprofen naproxen (41), and fenoprofen (see Analgesics, antipyretics, and antiinflammatory agents Salicylic acid and related compounds). [Pg.255]

The aluminum salts of dmgs are significant for two principal reasons reduction of dmg acidity in the stomach lining and reduction of undeskable flavor. Examples are the aluminum salts of glycine, acetylsahcyhc acid, ibuprofen, flurbiprofen, fenoprofen, flufenamic acid, oxaprozin, and a variety of aminobenzoic acid. Some of these compounds are Hsted in Table 2. [Pg.143]

Analgesic and antipyretic ibuprofen, 2-(p-isobutylphenyl)propionic acid, is converted to its monohydroxyaluminum salt by converting the acid to its sodium salt using dilute caustic, foUowed by concurrent addition of equimolar amounts of aluminum nitrate and sodium bicarbonate (84). AGP columns have wide appHcation for the direct separation of enantiomers of many different classes of dmgs, amines, acids, and nonprotolytic compounds (18,23). Acidic dmgs resolved include ibuprofen [15687-27-17, C 2H g02, ketoprofen [22071 -15 ] and naproxen [22204-53-17,... [Pg.99]

Lipase-catalyzed kinetic resolutions are often practical for the preparation of optically active pharmaceuticals (61). For example, suprofen [40828-46-4] (45), which is a nonsteroidal antiinflamatory dmg, can be resolved by Candida glindracea]i 2Lse in >95% ee at 49% conversion (61). Moreover, hpase-based processes for the resolution of naproxen [22204-53-1] and ibuprofen [15687-27-1] (61) have also been developed. [Pg.338]

Recently, Picciola et al. (81FES1037) prepared some 2iT-indazole derivatives containing a phenylalkanoic acid residue with potential antiinflammatory activity (693). M.G. 18755 (R = CHMeC02H, R = R = R = H) and its lysine salt, M.G. 18334, showed greater activity than ibuprofen, and the homologous butyric acid derivative M.G. 18860 showed good activity as a platelet aggregation inhibitor. [Pg.293]

The obtained analytical forms are used for quantitative determination of both Diclofenac natrium and Ibuprofen content in medicinal forms as well as for the testing uniformity of batching of the basic substance in tableted forms.
