The major problem in such conversions is the degradation of the branched carbon side-chain on C-17 which is present in all abundant steroids and lacking in all steroid hormones. The most important starting material used in industry today is diosgenin from the Mexican dioscorea plant. It is degraded by the method of Marker to 16-dehydropregnenolone in 45% total yield. This compound is a key substance in the production of several hormones with anabolic, catabolic, and sexual effects.

NaturaHy occurring and synthetic estrogens and androgens have been extensively and safely used to improve efficiency and carcass composition ia growing beef catde in the United States since the early 1950s. Several anaboHc steroid implants have been approved for use in beef catde in the United States, but only one, zeranol [55331-29-8] is approved for use in lambs. Anabolic steroids are not used for growth regulation in swine or poultry (see Steroids).

Classification of the anabolic steroids is based on chemical stmctures and associated actions. A review of the biosynthesis and metabolism of the naturally occurring estrogens and androgens is available (1). Names, descriptions, approval dates, and recommended doses of the commercial products are found in References 1, 8, and 9. Although steroids may be orally active, the FDA approved mode of adrninistration is the subcutaneous implant. Effective dose is lower with implant rather than oral adrninistration. 

FIAC also strongly inhibits HCMV and Epstein-Barr vims (EBV) in vitro the two vimses known not to induce a specific viral thymidine kinase for their repHcation. However, HCMV may stimulate cellular kinases that can anabolize FIAC to its 5 -triphosphate, which specifically inhibits the HCMV-encoded DNA polymerase. This selective activity suggests that FIAC should be evaluated against HCMV infections. FIAC-ttiphosphate incorporated into DNA has shown strong in vitro activity against the DNA polymerases of human hepatitis B vims (HBV) and of woodchuck hepatitis vims (WHV) (37).

In 1959 Clinton and coworkers reported the first synthesis of some pyrazole fused androstane derivatives and described their biological activity (B-76MI40404). Stanazolol (695) or 17-methyl-2iT-5o -androst-2-eno[3,2-c]pyrazol-17/3-ol was 10 times as active as 17a -methyltestosterone in improving nitrogen retention in rats (B-80MI40406), and its myotrophic activity was only twice that of 17a-methyltestosterone. It is used as an anabolic steroid with no lasting adverse side effects. 

Pharmacologically useful isoxazoles (B-82MI41600) include antibacterial sulfonamides (614), (615) and (616), semisynthetic penicillins (617), (618), (619) and (620), semisynthetic cephalosporin (621), anabolic steroid (622), the monoamine oxidase inhibitor (623) (used in psychotherapy), antiinflammatory agent (624) and antitumor agent (625). 

Farmed animals tend to be subjected to compounds that can be classified more easily as drugs. In some cases, animals may encounter compounds in nature that have considerable limitations in inclusion in the diets of farmed animals. An example is the inclusion of zearalenone (Figure 1) and its derivatives in animal diets these have anabolic and oestrogenic properties " and are permitted to be used in some areas of the world but are prohibited from use in others, such as European Union countries. Thus this compound and its reduced isomers (zearalenols, zeranols) improve growth rates in animals but can cause reproductive problems, especially in pigs. Ralgro, which is a commercially prepared derivative of zearalenone, is used as an ear implant in beef cattle as a growth promoter in...

The application of the Birch reduction to ethers of estradiol by A. J. Birch opened up the area of 19-norsteroids to intensive research. The major Birch reduction product is an enol ether which affords either a 3-keto-A -or a 3-keto-A -19-norsteroid depending upon the hydrolysis conditions. Various 19-norsteroids have been found to have useful clinical activity compounds (30), (31), and (32) are oral contraceptive agents and compound (33) has been used as an oral anabolic agent. Several of these compounds were prepared on an industrial scale for a number of years by the Birch reduction of estradiol derivatives. 

B-Homosteroids were first reported by Ringold in the course of an investigation of the relationship of structural changes to biological activity of anabolic agents. The Tiffeneau rearrangement sequence described earlier for the preparation of A-homosteroids was used to synthesize 17/ -hydroxy-B-homo-5a-androstan-3-one (56a).
