Moved] Cranberry fruit of Early Black cultivar was fractionated chromatographically and fractions were analyzed for flavonoid content. The effects of the flavonoid fractions and ursolic acid, an abundant triterpenoid in cranberry peel, were assessed in two models of colon cancer and one model of breast cancer. Clonogenic soft agar assays were used to determine the effect of these compounds on tumor colony formation in HCT-116, HT-29 and MCF-7 cells. MTT and trypan blue assays were performed to assess their ability to inhibit tumor cell proliferation. TUNEL assays were performed to assess apop-totic response to the cranberry compounds. The proanthocyanidins inhibited tumor colony formation in HCT-116 and HT-29 cells in a dose-dependent manner, with greater effect on the HCT-116 cell line. Ursolic acid strongly inhibited tumor colony formation in both colon cell lines. These compounds also decreased proliferation in all three tumor cell lines with the HCT-116 cell line most strongly affected. (150 words)...

Cell Lines. The human fetal intestinal cell line (FHS) was kindly supplied to us by Dr. Walter A. Nelson-Rees at the Naval Bioscience Laboratory, Oakland, California. The human colonic cell line, SKCO-1 developed by Drs. G. Trempe and L.F. Olds, was obtained from Dr. Jorgen Fogh, Sloan Kettering Institute, Rye, New York. The HT-29 cell line was developed and obtained from Dr. J. Fogh. SW-480 and SW-620 were developed at Scott and White Clinic in Temple, Texas and were obtained from Col. A. Liebovitz. 

The concentration of sodium butyrate was observed to have a differential effect on cell growth in colonic cell lines. After culturing for 8 days with 5 mM sodium butyrate, the cell protein per flask of the SCKO-1 line was decreased to less than 10% of the control cultures. In the SW-620 culture,cell protein per...

For assessing the absorption potential of specific drug candidates or conducting studies evaluating correlations between drug structure and transport, in vitro models may provide the best approach. Both the tissue-diffusion cell systems and cultured colon cell lines have proved to be particularly useful. 

Palominol. Corey has reported the application of an intramolecular titanium-mediated pinacol coupling reaction to the synthesis of a 15-membered ring, en route to palominol, a marine diterpenoid that displays cytotoxicity toward the human colon cell line (Eq. 3.10) [28]. Slow addition of the keto aldehyde (32 h) to the titanium reagent furnished the cyclized product in 53% yield as a mixture of diastereomers (2.1 1). 

Intestinal tract is the portal of entry for many xenobiotics such as orally administered pharmaceuticals, dietary constituents, and contaminants and even for ophthalmic drugs applied on the cornea and drained via uveoscleral tube to nasopharynx and gastrointestinal tract. Obviously it is difficult to find proper representative cells for the whole length of intestinal tract, but if the focus is on the absorption and metabolism, the colonic cell line Caco-2 has been used for decades and its characterization provides a good example of the difficulties associated with extrahepatic cell lines. Principally in food research area, several human cell lines, e.g., HIEC-6, H4, and H4-1, have been employed for bioaccessibility, absorption, and biotransformation studies [28, 29], but their application in toxicological research has been relatively unexplored.
