Disintegration, tablet

The crospovidones are easily compressed when anhydrous but readily regain their form upon exposure to moisture. This is an ideal situation for use in pharmaceutical tablet disintegration and they have found commercial appHcation in this technology. PVP strongly interacts with polyphenols, the crospovidones can readily remove them from beer, preventing subsequent interaction with beer proteins and the resulting formation of haze. The resin can be recovered and regenerated with dilute caustic. 

Tablet-disintegration tests consist of cychcal immersion in a suitable dissolving fluid of pharmaceutical tablets contained in a basket. Acceptable tablets disintegrate completely by the end of the specified test period United State.s Fharmacopeia, 17th rev.. Mack Pub. Co., Easton, Pa., 1965, p. 919).

Sodium alcohol sulfates are also used in the formulation of synthetic soaps and paste hand cleaners, commonly together with other surfactants and as tablet disintegrators in the case of sodium dodecyl sulfate. Sodium, but preferably ammonium and alkanolamine salts, is also used in liquid soaps.

Garlic supplements - powder tablets or capsules, steam-distilled oil, vegetable oil macerate extract, or extract aged in dilute alcohol - are widely available and are taken by millions. Since the active principle, allicin, is not present in garlic bulb, the supplements rely on the presence of precursor alliin and enzyme alliinase. In tests on 24 commercial brands of enteric-coated tablets, all except one gave low dissolution allicin release 83% of the brands released less than 15% of their potential allicin.78,79 Relevant factors were impaired enzyme activity caused by excipients and slow tablet disintegration. Studies have shown that the pore size distribution can influence liquid penetration into tablets [28,29,52]. The accessibility of water vapor to tablet components has been related to chemical stability, especially in the case of hydrolyzable drugs [28]. Solvent penetration, which can affect tablet disintegration and dissolution, has been related to pore size [52].

Although water penetration is a neeessary first step for disintegration, swelling is probably the most widely aceepted mechanism of action for tablet disintegrants. Indeed, most disintegrants do swell to some extent, but the variability of this property between disintegrants reduces its plausibility as a sole meehanism.

Immediately upon opening the blister, using dry hands, remove the tablet and place it in the mouth. Tablet disintegration occurs rapidly in saliva so it can be easily swallowed with or without water.
