Since the catabolism of Ig is similar in both lines, th. difference of Ig serum levels is due to an impairment of the general Ig synthesis in Low responders. Mucosal and systemic antibody responses were measured following oral (OR) and SQ immunization by a nanoparticulate-formulated protein. We have examined the immunosuppressive effect of IL-2-toxin in a murine DTH model. BALB/c mice were immunized by subcutaneous injection of the hapten trinitrobenzenesulfonic acid (TNBS). Seven days following immunization, mice were challenged with a 25 pi injection of TNBS into the right footpad. After 24 hr, bilateral footpad thickness was measured with a micrometer in order to assess the degree of the immune response. The quality of mRNA is important in generating recombinant antibodies. It is particularly important to ensure minimal ribosomal RNA contamination when cDNA is random primed for Ubraiy construction to reduce the proportion of clones which represent non-mRNA species. The spleen mRNA of immunized mice is presmnably derived mainly from plasma cells, as the level of Ig mRNA in these cells is up to 1000-fold greater than in resting B cells. For spleens, extraction of total RNA followed ly mRNA isolation on oligo(dT)-cellulose is recommended in order to effectively process the large number of cells present in a spleen. Previous studies in our laboratory have utilized the New Zealand Black (NZB) mouse strain as a model of autoimmune disease (Siegel and Morton, 1970 Morton and Siegel, 1980). We now report results of experiments which suggest a possible role for vitamin C in autoimmunity in the NZB mouse. Old NZB mice exhibit lower spleen vitamin C levels than do non--auto immune strains (DBA/a and BALB/c), as is shown in Table II. In each instance, NZB mice showed significantly lower levels of vitamin C in the spleen when compared to non-autoimmune mice, suggesting a possible relationship between depressed spleen vitamin C levels and autoimmunity. [Pg.11]
