Specialized Methods

There are, of course, other methods for the preparation of amino acids. Often, these methods are specific to one system and although they can sometimes be used for other syntheses, there is no obvious or general linkage to one of the categories listed above. Several such strategies are collected into this section. [Pg.46]

There are several synthetic approaches that rely on a molecular rearrangement to incorporate the appropriate functionality. A classical reaction is the Curtius rearrangement,i43 in which an azido ketone rearranges to an isocyanate. Subsequent hydrolysis converts the isocyanate to an amine. This approach can be applied to the synthesis of amino acids. When 2,2,3,3-tetramethylbutanedioic acid (L249) was [Pg.46]

Modifications of this basic approach include functionalization and rearrangement of trichloroacetamide derivatives. Dienoic amino acids are possible, as when 1.255 was converted to 1.256. Thermal rearrangement led to 1.257 and deprotection followed by oxidation gave 3-(N-Boc amino)hexa-2E,4-dienoic acid, 1.258. [Pg.47]

There are several other interesting methods that convert alkenes to amino acid derivatives. All include somewhat novel oxidative methods. Ruthenium complex 1.259, for example, was converted to methyl 4-(N,N-dimethylamino)-2-methylbut-anoate (,1.260). In addition, methyl 2-ethyl-4-(N,N-dimethylamino)butanoale [Pg.47]

Silverman modified this approach by condensing lithio t-butyl acetate with a-fluoro-acetate to give 1.266. Subsequent reduction of the enamine moiety gave 1.265.  [Pg.49]
