Robot System for Ultrasonic Examination of Complex Geometries. 

The cost of the vehicle to move the accelerator, and temporary shielding, if needed, have not been included in Table 14. The vehicle houses the control console, video system, and robotic controls. A trailer would carry the main robotic system, the electronic accelerator, magnetron, and the supplies. The cost for such a vehicle and trailer is estimated at 100,000. 

A regularly formed crystal of reasonable size (typically >500 pm in each dimension) is required for X-ray diffraction. Samples of pure protein are screened against a matrix of buffers, additives, or precipitants for conditions under which they form crystals. This can require many thousands of trials and has benefited from increased automation over the past five years. Most large crystallographic laboratories now have robotics systems, and the most sophisticated also automate the visualization of the crystallization experiments, to monitor the appearance of crystalline material. Such developments [e.g., Ref. 1] are adding computer visualization and pattern recognition to the informatics requirements. 

In the last several years, on-line extraction systems have become a popular way to deal with the analysis of large numbers of water samples. Vacuum manifolds and computerized SPE stations were all considered to be off-line systems, i.e., the tubes had to be placed in the system rack and the sample eluate collected in a test-tube or other appropriate vessel. Then, the eluted sample had to be collected and the extract concentrated and eventually transferred to an autosampler vial for instrumental analyses. Robotics systems were designed to aid in these steps of sample preparation, but some manual sample manipulation was still required. Operation and programming of the robotic system could be cumbersome and time consuming when changing methods.

A 10 mM ionic strength universal buffer mixture, consisting of Good zwitterio-nic buffers, [174] and other components (but free of phosphate and boric acid), is used in the pION apparatus [116,556], The 5-pKa mixture produces a linear response to the addition of base titrant in the pH 3-10 interval, as indicated in Fig. 7.53. The robotic system uses the universal buffer solution for all applications, automatically adjusting the pH with the addition of a standardized KOH solution. The robotic system uses a built-in titrator to standardize the pH mapping operation. 

The most recent extension of instrument automation has come with the availability of practical laboratory robotics systems. These systems can be as easy to implement as the personal computer data system and extend automation beyond control, data collection and... 

Directed evolution relies on the analysis of large numbers of clones to enable the discovery of rare variants with unproved function. In order to analyze these large libraries, methods of screening or selection have been developed, many of which use specialized equipment or automation. These range from the use of multichannel pipettes, all the way up to robotics, depending on the level of investment [59]. Specialized robotic systems are available to perform tasks such as colony picking, cell culture, protein purification, and cell-based assays. 

As already mentioned above, a different strategy to achieve high throughput in microwave-assisted reactions can be realized by performing automated sequential microwave synthesis in monomode microwave reactors. Since it is currently not feasible to have more than one reaction vessel in a monomode microwave cavity, a robotic system has been integrated into a platform that moves individual reaction... 

Many sensors have to be integrated into such mobile robotic systems, with a major share of miniaturized, low-power microsystems. Self-guiding features are used in such robots, as well as autocharger docking system features. 

The simplest technique is the use of the 96-well collection plate format (analogous to the format used in SPE) in conjunction with a liquid handling robotic system it follows the same principle as bulk scale LLE. However, immobilization of the aqueous plasma sample on an inert solid support medium packed in a cartridge or in the individual wells of a 96-well plate and percolating a water-immiscible organic solvent to extract the analyte from this medium evoked significant enthusiasm from the pharmaceutical industry. 

Laser nephelometry is based on measuring the turbidity of an aqueous medium after adding a fixed amount of solution of a compound in dimethyl sulfoxide (DMSO).41 Dehring et al.42 described an automated robotic system with laser-based nephelometry for high-throughput kinetic aqueous... 

MS has recently been used to measure compounds with significant levels of impurities and solubilities below the quantitation limits of other methods. Guo et al.46 described the use of LC/MS for solubility measurements in buffer solutions in a 96-well plate. Fligge et al.47 discussed an automated high-throughput method for classification of compound solubility. They integrated a Tecan robotic system for sample preparation in 384-well plates and fast LC/MS for concentration measurement. This approach is limited by LC/MS throughput. 

Any decision to establish automated or robotic systems must carefully consider prerequisites such as the annual numbers of samples to be processed to achieve an acceptable cost-to-benefit ratio. Late phase development stability studies may benefit from fully automated systems based on the enormous numbers of samples to be analyzed for each stability time point. The use of automated systems in manufacturing quality control is now required due to the sheer number of samples to be... 

For these reasons, researchers have recently focused on developing faster robotic systems and more sensitive analytical metabolite identification tools [5-9]. However, such techniques are usually resource demanding, consuming a considerable amount of compound, and cannot be used before compound synthesis. Also, because of the increasing number of potential candidates, experimental metabolite identification remains a huge challenge.
