Analytical Methods. The samples were analyzed by standard AACC (21) procedures for moisture (air-oven method), protein (Method 46-13), crude fat (Method 30-25), crude fiber (Method 32-10), insoluble dietary fiber (Method 32-20) and ash (600°C, 3 hr). Starch content was determined by the polarimeter method (Method 76-20) and total sugars by Method 80-60. Color characteristics of the dried products were evaluated with the Hunter Color Difference Meter. [Pg.183]

What follows is not an exhaustive or up-to-the-minute survey of the methods available for protein quantitation, but a practical guide to selecting the appropriate assay for each stage of drug development. A case study further illustrates the application of the standard protein methods to the drug development process. The reader is referred to reviews on the topic for further details.12... [Pg.15]

Bilirubin effects depend on the method used for analysis. Interferences in direct serum protein methods are observed at bilirubin levels greater than 5 mg/100 ml (K7). A sample containing 20 mg of bilirubin per 100 ml increased the apparent total protein by 0.2 g/100 ml. Concentrations of bilirubin as high as 20 mg/ml do not effect albumin assays using bromocresol green binding (D12), but have a marked effect on these assays when [2-(p-hydroxyphenylazo)-benzoic acid] (HABA) dye is used (A7b). [Pg.7]

During this process some key questions are (1) Does the DNA sequence inserted into the expression vector contain all the sequences essential for the target protein (2) Is the messenger RNA expressed at the predicted length and produced in high enough levels (3) Does the recombinant protein react with antibody (4) Is there evidence of functional activity of the recombinant protein Methods for answering these questions are listed in Table 4.4. [Pg.46]

The discussion of activity from X-ray data in conjunction with kinetic data is also difficult because, apart from considerations of dynamics, these techniques do not provide the essential knowledge about the energy states of given atoms or groups. It is necessary to inspect the electronic structure of at least certain regions of the protein. Methods exist for this inspection, and these include electron paramagnetic resonance, ultraviolet, circular dichroism, Raman and Mossbauer spectroscopies. The full understanding of activity can only come when the information derived from all available methods is assimilated and rationalized. [Pg.56]
