Recombination occurs within a common core sequence of 15 base pairs. The excision reaction requires the X Xis protein in addition to Int and IHF. Figure 4.5. Vaccines made with recombinant proteins marshal an immune response to specific viruses to treat or prevent disease. Vaccines to prevent infection with the liver-damaging hepatitis viruses (Twinrix , Recombivax HB , and Engerix-B ) include a recombinant version of a hepatitis virus B protein. Recombinant hepatitis B protein is also included in some combination vaccines given to children. Comvax also includes a recombinant flu virus protein. [Pg.77]

For the purpose of synthesizing flavor peptides or proteins in large scale, we developed "protein recombination method" and "enzymatic synthesis using chemically modified enzyme". "Protein recombination method" was applied to the synthesis of C-terminal portion of p-casein and its analog. Chymotrypsin was chemically modified by Z-DSP in aqueous solution. It was stable for organic solvents. Using this modified enzyme, we succeeded in the synfiiesis of Inverted-Aspartame-Type Sweetener "Ac-Phe-Lys-OH" in one step. [Pg.149]

Today, it is well-known that peptides or proteins exhibit various kinds of taste. Our group has been researching on the relationship between taste and structure of peptides, BPIa (Bitter peptide la, Arg-Gly-Pro-Pro-Phe-Ile-Val) (7 as a bitter peptide, Om-p-Ala-HCl (OBA), Om-Tau-HCl as salty peptides(2j, and "Inverted-Aspartame-Type Sweetener" (Ac-Phe-Lys-OH) as a sweet peptide(5). The relationship between taste and chemical structure was partly made clear. Since commercial demand for these flavor peptides is increasing, we need to develop new synthetic methods which can prepare these peptides in large scale. We developed the following two methods (1) protein recombination method as a chemical method, (2) enzymatic synthesis using chemically modified enzyme as a biochemical method. [Pg.149]

A number of protocols will be described here that have been used successfully with both rat (Y3 and IR983F) and mouse (SP2/0) myelomas to generate MAbs to cellular proteins, recombinant proteins, or peptides based on cDNA sequences. The formation of beads is a two-step process based on the cold gelation of whey proteins in the presence of divalent cations, such as Ca2+ [67], Briefly, the whey protein isolate (WPI) solution (10% w/v in deionized water) was (i) adjusted at pH 7 to favor the apparition of negative charges implied in ionic bounds with Ca2+ ions and (ii) heated (80 °C, 45 min) to denaturate the proteins. Recombinant cells in the beginning of their stationnary growth phase were suspended in a sterile solution of... [Pg.580]
