Internal tissue breakdown in melons such as Galia and Cantaloupe occurs around the seedbed and eventually results in collapse of the seed bed into a mixture of liquid, seeds and tissue pieces. Unfortunately this disorder does not affect the skin or hardness of the outer layers so cannot be detected by visual inspection. Preliminary reports from the Volcani Center in Israel have shown that the tissue breakdown results in a small increase in T2 from, typically, 35 to 45 ms and a corresponding increase in signal intensity in spin-echo projection images acquired with a 4.7 T (200 MHz) imaging magnet. There is therefore a reasonable possibility of being able to detect this disorder by on-line projection MRI. [Pg.97]

General Observations. The chain and most of the associated side chains are not well defined in the following regions residues 2, 65-72, and 119-123. The chain is very poorly defined or not visible at all in regions residues 1, 18-20, 21-23, and 124. Except for the small peptide loop, these regions are all associated with the chain ends. Cursory inspection of the sums of the electron densities in the backbone and /3-carbon positions shown residues 3 through 9 to be systematically low, and preliminary calculations show that a simple shift of these residues by 0.6 A in the crystallographic z direction would bring these sums into line with those for the remainder of the structure. [Pg.665]

Detection by Fluorescence. The detection test is the ultraviolet light-activated yellow fluorescence which is induced only after spraying with 70% aqueous sulfuric acid (2). A preliminary inspection under ultraviolet light eliminates substances with non-sulfuric acid-induced ultraviolet fluorescence. In practice, the restriction to yellow eliminates many predominantly blue components in a plant extract chromatogram. Some yellow fluorescent components have been observed on occasion from plant sources without spraying, and are thus eliminated.
