Synthesis of peptides by methods of biogenetic engineering transcends the boundaries of peptide chemistry the method is generally applied for the production of proteins. Nevertheless, it seemed to be proper to conclude the history of insulin research with the presentation in a nutshell of the preparation of the hormone through the processes of biotechnology. 

Several syntheses of ACTH were reported in the following years. Of these the efficient construction of the molecule by Bajusz and his associates [112] is mentioned here. SoUd phase synthesis of the hormone on a commercial scale has been attempted but it is more practical at this time to manufacture ACTH in solution.

In spite of the conspicuous homology displayed in the MEHFRWG sequence )S-MSH is less closely related to ACTH than a-MSH. Difficulties encountered in the synthesis of jS-MSH prompted Schwyzer to seek the new tactical approach discussed in connection with the synthesis of ACTH.

Melatonin [73-31-4] C 2H N202 (31) has marked effects on circadian rhythm (11). Novel ligands for melatonin receptors such as (32) (12), C2yH2gN202, have affinities in the range of 10 Af, and have potential use as therapeutic agents in the treatment of the sleep disorders associated with jet lag. Such agents may also be usehil in the treatment of seasonal affective disorder (SAD), the depression associated with the winter months. SRIF acts as an excitatory neuromodulator in the CNS inhibiting the release of TRH, corticotropin-releasing hormone (CRH), growth hormone releasing factor (GHRH), and NE. It produces general arousal and hypotension. It inhibits the release of a number of peptides and modulators in the GI tract.

CRH (Corticotropin releasing hormone) is expressed in the nucleus paraventricularis of the hypothalamus and drives the stress hormone system by activating synthesis and release of corticotropin at the pituitary and in turn corticosteroid from the adrenal cortex. CRH is also expressed at many other brain locations not involved in neuroendocrine regulation, e.g. the prefrontal cortex and the amygdala. Preclinical studies have shown that CRH also coordinates the behavioral adaptation to stress (e.g. anxiety, loss of appetite, decreased sleepiness, autonomic changes, loss of libido). 

The paraventricular nucleus in the hypothalamus is located adjacent to the third ventricle and has been identified as a satiety center. Neurons in the paraventricular nucleus produce neuropeptides which inhibit feeding when injected into the brain (thyrotropinreleasing hormone (TRH), corticotropin-releasing hormone (CRH), oxytocin).

Corticotropin (ACTH) is an anterior pituitary hormone that stimulates the adrenal cortex to produce and secrete adrenocortical hormones, primarily the glucocorticoids. 

Corticotropin is used for diagnostic testing of adrenocortical function. This drug may also be used for the management of acute exacerbations of multiple sclerosis, nonsuppurative thyroiditis, and hypercalcemia associated with cancer. It is also used as an anti-inflammatory and immunosuppressant drug when conventional glucocorticoid therapy lias not been effective (see Display 50-1). 

ACTH is contraindicated in patients with adrenocortical insufficiency or hyperfunction, allergy to pork or pork products (corticotropin is obtained from porcine...
