Binding affinities of the new flexible synthetic glycocluster toward type-1-piliated E. coli were evaluated by HAI and bladder-binding assay (BBA). The results indicated that, with an inhibition of bacterial bladder-cell binding at 12 nM ( 6000- and 64-fold lower than mannose and 59, respectively), the tetravalent compound 60 is currently one of the most promising antiadhesive drugs under development for the treatment of urinary tract infections. [Pg.190]

Cranberries have been used traditionally for the treatment and prevention of urinary tract infections. Their effectiveness was demonstrated by a randomized, double-blind placebo-controlled trial (Avom et ah, 1994). Escherichia coli are the principal bacterial species responsible for urinary tract infection. The consumption of cranberry juice reduced the adherence of E. coli to the uroepithelial bladder cells in healthy human volunteers (Di Martino et ah, 2006). A-type procyanidin dimers and trimers that were isolated from cranberries were found to inhibit the adherence of uropathogenic E. Coli, whereas (—)-epicatechin and a B-type dimer were not effective (Foo et ah, 2000). [Pg.251]

The infrared spectrum was taken of the entire cell, whereas the Raman spectrum was sampled at a small spot within the cytoplasm. Both spectra are presented in arbitrary intensity units, along the same wavenumber axis. Because most of the broccoli ITCs ingested by humans are excreted in urine and storage of urine in the bladder may allow a degree of dissociation of dithiocarbamates to free ITCs, high intracellular accumulation of ITCs in bladder cells may be more readily achieved. Since the transitional epithelia that line the inner surface of the bladder are the... [Pg.124]

Biofllms are complex bacterial commtmities which are encased by a protective exopolysaccharide (EPS) matrix that helps the bacteria thrive in hostile environmental conditions and reduces the efficacy of antibiotics by up to 100-fold relative to planktonic cells/ Briefly, the formation of biofllms can be described as a multistep process. The planktonic bacteria are first attached by strong association of the adherent cells to the surface such as respiratory mucosa and bladder cells in case of lung and urinary tract infections, respectively. Upon attachment, the bacteria multiply to form microcolonies. These microcolonies develop into well-defined mushroom-like three-dimensional structures and eventually produce the EPS coating around them. At times, the biofilm matrix breaks and the bacterial cells disperse, which leads to a spread in infection (Figure 16). ... [Pg.280]

Aiming for a diagnostic urine test for transitional cell carcinoma (TCC, 95% of total cases), 94 samples and controls were analyzed. Of some 70 differentially expressed proteins and polypeptides in the 2-150 kDa mass range, 5 were preferentially expressed in TCC, at 3353 Da, 9495,44.6,100.120, and 133.190 kDa. The 3.3 kDa protein, also detected in microdissected bladder cells, was identified, by SELDI immunoassay and database search, as a member of the human defensin family. The diagnostic sensitivity of the combined markers was 78% compared to 33% of cytologic approaches [97]. [Pg.390]
