Walker tumor

Colquhoun and Schumacher [98] have shown that y-linolcnic acid and eicosapentaenoic acid, which inhibit Walker tumor growth in vivo, decreased proliferation and apoptotic index in these cells. Development of apoptosis was characterized by the enhancement of the formation of reactive oxygen species and products of lipid peroxidation and was accompanied by a decrease in the activities of mitochondrial complexes I, III, and IV, and the release of cytochrome c and caspase 3-like activation of DNA fragmentation. Earlier, a similar apoptotic mechanism of antitumor activity has been shown for the flavonoid quercetin [99], Kamp et al. [100] suggested that the asbestos-induced apoptosis in alveolar epithelial cells was mediated by iron-derived oxygen species, although authors did not hypothesize about the nature of these species (hydroxyl radicals, hydrogen peroxide, or iron complexes ). [Pg.756]

Busch and his associates [18-21] attempted to isolate nucleoli from either Walker tumors or liver treated with thioacetamide. They identified the nucleoli with the aid of azure and claim to have obtained, after sonication of the nuclei followed by differential centrifugation, a nucleolar preparation 90-95% free of contamination. The preparation is rich in DNA, relatively poor in RNA but because the yield in nucleoli is low, their nucleolar preparation can hardly be considered representative of the original nuclear preparation. [Pg.78]

The first data on the effects of tamoxifen on uterine leiomyomas were obtained in a rat model. They showed that tamoxifen increased tumor latency and decreased tumor size (Howe et al. 1995). These findings were confirmed more recently by Walker et al. (2000). In the same period, moreover, several case reports were published showing an increase in uterine leiomyoma dimensions following tamoxifen administration (Dilts et al. 1992 Leo et al. 1994 Ugwumadu et al. 1994). [Pg.304]

B16 melanoma, Walker sarcoma, and M5076 forming liver metastasis have been used in the preclinical evaluation of HPMA copolymer-adriamydn conjugates [36]. Other tumors useful for secondary screening are MS-2 sarcoma, NMU-1 murine lung adenocarcinoma, and murine adenocarcinoma Colon 26. These have been used by Zunino et al. [147] to determine the activity of poly (carboxylic acid) immobilized anthracyclines. Mice inoculated intramuscularly with Lewis lung carcinoma have been used by Pratesi et al. [215] to assess the effect of a poly-L-aspartic acid/doxorubicin conjugate. [Pg.92]

In a similar study by Yang and Huang (1996), melanoma tumors in mice were in vivo transfected by the direct intratumoral administration of naked pDNA, resulting in reporter gene expression for up to ten days. In another study, Walker 256 carcinoma tumors in rats were found to express CAT after intratumoral injection of naked pDNA encoding CAT (Nomura et al., 1997). More recently electroporation has been used to deliver naked pDNA to tumors (described further in the following section). [Pg.264]
