For determination of total fatty acids lipids must be split by alkaline hydrolysis. Hydrolysis of lipids from purified lipid extracts is preferable to hydrolysis of tissues or serum directly. After extraction of unsaponifiable matter fatty acids are recovered from their soaps by treatment with acids and can subsequently be assayed by titration. During hydrolysis high concentrations of alkali must be avoided to prevent isomerization of unsaturated fatty acids. A suitable method for quantitation of plasma total fatty acids was described by Albrink (1959/1960). [Pg.201]

Free fatty acids occur only in small amounts in biological materials. They can be recovered from ether or petroleum ether extracts by extraction with diluted aqueous or alcoholic alkaline solutions as the corresponding alkaline salts and reconverted to free acids by treatment with acids. The use of diluted alkaline solutions prevents hydrolytic cleavage of glycerides and glycerophospholipids. Free fatty acids may also be isolated by adsorption to basic ion exchange resins (Carlson and Wadstrom 1958, Hornstein et al. 1960). Because of a tendency of polyunsaturated fatty acids to isomerization Biegler et al. (1960) recommend the use of weak basic adsorbents. After isolation free fatty acids are assayed by titration. [Pg.201]

A convenient method for quantitation of free fatty acids in plasma, incubation mixtures, tissue homogenates etc. was introduced by Dole (1956), according to which the free fatty acids are extracted directly with an isopropanol-heptane mixture from acidified samples. Further addition of heptane and water results in formation of a two-phase system. Aliquots of the upper heptane phase containing the free fatty acids are titrated with a dilute solution of aqueous sodium hydroxide. [Pg.201]

For elimination of dicarbonic acids and phospholipids which interfere with titration washing of the final heptane extracts with dilute sulfuric acid is recommended (Trout et al. 1960, Friedberg et al. 1960). For the same reasons Gordon and Cherkes (1956) and Gordon et al. (1957) used extraction of lyophil-ized plasma with isooctane and acetic acid. Lyophilization is unnecessary if extraction is performed with isooctane-acetic acid anhydride-acetic acid-sulfuric acid mixture (Shafrir 1958, Shafrir and Steinberg 1960). [Pg.201]
