The effects of charge proximity on CD complexation behavior were evaluated (Fig. 7) by studying the complexation of two steroids by the sulfonate, sulfopropyl ether (SPE), and sulfobutyl ether (SEE) derivative.Electronic effects seem to be more of a factor than steric effects because even when only one sulfonate substituent is attached at the 6-position, (6-SAl-p-CD) the derivative loses its complexation capability. The binding constant for testosterone is only 64 for 6-SAl-p-CD versus 17,800 for the neutral p-CD. The attachment of a single negative charge close to the CD cavity appears to disrupt the thermodynamics driving the complexation. [Pg.678]

One approach to introduce an anionic moiety onto dextran is sulfopropylation. In combination with a hydrophobic modification, sulfopropylation was utilised for manufacture of anionic amphiphilic dextran ethers. The derivatives were synthesised in two steps. Dextran was reacted with 1,2-epoxy-3-phenoxy propane in aqueous NaOH solution (Fig. 35) or in DMSO using TBAH instead of NaOH. The subsequent step is the conversion with 1,3-propane sultone in DMSO (Fig. 37). [Pg.250]

The sulfopropyl and sulfobutyl ether derivatives " have been further evaluated for their complexation with testosterone and progesterone (Fig. 7). Even though increasing the DS should produce more steric hindrance to complexation, the mono, tetra, and hepta substituted sulfobutyl ether (SBEl, 4, 7) derivatives all displayed comparable binding abilities for the steroids and the strength of binding was similar to that observed for p-CD. The SBE substituent behaves similar to that proposed for the undecyl sulfate CD, however, complexation with the SBE-p-CDs involves the CD cavity, as well as the hydrophobic butyl side arms. [Pg.677]
