SB225002 was among the first small-molecule chemokine receptor antagonists described, and it has been used in animal models to inhibit IL-8-induced neutrophil margination (White et al, 1998). A modification of this compound, with an IC50 for blockade of CXCL8 binding of 9.3 nM, attenuates leukocyte infiltration into the synovium in models of acute and chronic arthritis in rabbits (Podolin et al, 2002). [Pg.336]

In vitro studies have documented that activation of leukocytes by ANCAs can release factors that are injurious to endothelial cells (52-59). Endothelial injury by ANCA-activated leukocytes requires interplay between multiple cytokine and adhesion molecule ligands and their receptors in the microenvironment at the site of vasculitis (55-59). Overall, in vitro activation of neutrophils by ANCA IgG causes neutrophil adherence to endothelial cells, diapedesis through endothelial monolayers, and release of toxic factors that kill endothelial cells (Fig. 1). In the very early phases of the acute inflammatory response most of the cells invading the damaged area are polymorphonuclear neutrophils, also denoted as PMNs, which serve as initial line of defense and source of proinflammatory cytokines. The CSFs should not be used routinely for treatment of febrile neutropenia in conjunction with antimicrobial therapy.5 However, the use of CSFs in certain high-risk patients with hypotension, documented fungal infection, pneumonia, or sepsis is reasonable. These cells are crucial in fighting viral infections as part of the early innate response. Severe systemic viral infections, specifically herpes virus, may result from a lack or malfunction of NK cells. Patients infected with human immunodeficiency virus (HIV) have low numbers of NK cells. These cells also play a role in killing tumors. Patients with Chediak-Higashi syndrome have an increased risk of lymphomas, and this disease is associated with impaired NK cells, macrophages and neutrophils. A number of cytokines including IL-2, IL-12, IL-15, IL-21, IFN-a and IFN-(3 induce NK cells, which results in their proliferation, margination, cytokine production and cytotoxicity. [Pg.13]

During the past two decades, essentiality of zinc for man has been established. Deficiency of zinc in man due to nutritional factors and several dls-seased states, has been recognized. A marginal deficiency of zinc appears to be prevalent in many segments of population in developed countries and more severe deficiencies are widespread in many parts of the world. In our experimental human model, a marginal deficiency of zinc was induced by dietary means. Loss of body weight (less than 10% in six months on zinc restricted diet), testicular hypofunction, hyperammonemia and a decrease in plasma, urinary and neutrophil zinc concentration were observed. Changes in zinc dependent enzymes such as deoxythymldine kinase in newly synthesized connective tissue and plasma alkaline phosphatase were also observed as a result of zinc restriction and repletion in our model. [Pg.1]

The palpebral conjunctiva is composed of nonkera-tinized stratified epithelium that decreases in thickness as it proceeds from the eyelid margin. Many mucin-secreting goblet cells are located near the fornix. The epithelium overlies the substantia propria, which consists of delicate connective tissue and blood vessels.
