Endothelial dysfunction increases adhesiveness and permeability of the endotheMum for platelets and leukocytes. Infiltrations involve monocytes and T cells. MAbs make excellent tools with which to begin elucidating the relationship between chemokine, receptor, leukocyte recruitment, and disease. Flow cytometric analysis of chemokine receptor expression on peripheral blood leukocytes together with immunohistochemical examination of chemokine receptor expression on leukocytes recruited to inflammatory lesions may be used to correlate specific receptors with disease associated leukocyte infiltrates. 

The slides are checked for inflammatory signs, such as edema, leukocytic infiltration or foci of coagulative necrosis. 

Human CEC cultured on filter inserts can be used to measure brain penetration of drugs to make in vitro-in vivo correlations. They can further be used to study for example brain inflammation that has been implicated in the development of brain edema and secondary brain damage in ischemia and trauma. Zhang et al. (2000) studied leukocyte infiltration across the blood-brain barrier and showed the expression of adhesion molecules and leukocyte chemoattractants under ishemia in vitro conditions.

Leukocyte infiltration across the blood brain bariier after ischemic stroke is dependent on the expression of adhesion molecules on leukocytes and endothelial cells.
