The first description of an occupational cancer was made by a British surgeon, Percival Potts in 1775. This was cancer of the scrotum in young chimney sweeps in the 18th century in Britain. Many young boys, who were small enough to climb up chimneys, were employed to remove the soot. They probably worked whilst clad in very few clothes so that there would be frequent damage to the scrotum which, with direct exposure to soot, could lead to the development of cancer, which was known as chimney sweep s cancer or epithelioma of the scrotum. 

Cancer was first recognized as an occupational hazard in 1775 when the prevalence of scrotal cancer among London chimney sweeps was noticed. The chemical origin of this form of cancer was not universally accepted until 1922 when it was demonstrated that tumors could be induced on mouse skin using an etheral soot extract (60). Skin cancer was also noted to be an occupational hazard of workers exposed to pitch dust in the coal-briquetting industry (61) and to workers exposed to cmde tar (62). In 1915, tumors were produced in rabbits ears by prolonged application of cmde coal tar (63). Some 38 cases per year have been reported of cutaneous epithelioma in the tar distilling industry over a 25-year period up to 1945 (64). 

Joos B has reported that ointments centaming 0.1-0.4% of a Tuixture of propiolactone and aqueous albumin 30%) gi-ve excellent cosmetic results with regeneration of the skin. However, if one grants that Complete hydrolysis does not occur rapidly, there are objections to its cosmetic application in vitro propiolactone destroys certain enzymes of skin samples stored for use as grafts ° it is a vesicant (seotkm IX.l..d) and in addition osjtnnogenk. It initiates epitheliomas in rats when iq lied locally. 

Topical fluorouracil has been associated with allergic contact dermatitis in patients with actinic keratosis and basal cell epitheliomas (116). Topical fluorouracil ointment (5%) was reported to exacerbate dermatitis when it was mistakenly given instead of fluocinonide ointment (117).

Cannabinoids have been shown to induce cell cycle arrest in breast carcinoma (De Petrocellis et al. 1998), prostate carcinoma (Melck et al. 2000) and thyroid epithelioma cells (Bifulco et al. 2001). In breast carcinoma cells this has been ascribed to the inhibition of adenylyl cyclase and the cAMP/protein kinase A (PKA) pathway (Table 1). PKA phosphorylates and inhibits Raf-1, so cannabinoids prevent the inhibition of Raf-1 and induce prolonged activation of the Raf-l/MEK/ERK signalling cascade (Melck et al 1999). Cannabinoid-induced inhibition of thyroid epithelioma cell proliferation has been attributed to the induction of the cyclin-dependent kinase inhibitor p27 P (Portella et al. 2003).

Administration of condensed fumes from both types of coal tar pitches and both types of asphalt resulted in benign tumors (papillomas, kerato-acanthomas, fibromas, and unclassified benign epitheliomas) and malignant tumors such as squamous cell carcinoma and fibrosarcomas (seen more often in C3H/HeJ mice) (Niemeier et al. 1988). Anthramycin (95), produced by Streptomyces refuineus [171], is active in vivo against sarcome 180, Ehrlich solid and Ehrlich ascites carcinomas, Waker 256 carcinosarcoma and human epithelioma no. 3 [172]. Badger et al. (140) reported that oral administration to 10 mice of 5 mg/week/mouse of dibenz[a,/j]acridine resulted in one mouse with an epithelioma and papilloma of the forestomach another mouse showed stomach papillomas. Duration of the experiment was 627 days. Similar administration of dibenz[a,y]acridine (10 mice, 5 mg/week/mouse) gave no tumors in 572 days. In their introduction, the authors stated ... 

The specific effect of beryllium in DNA metabolism may be highly relevant in the elucidation of its experimentally demonstrated carcinogenic action.
