Epigenetics

Epigenetics can be defined as the study of how heritable changes in phenotype are transmitted without changes in the underlying DNA sequence, as defined by the four canonical bases, A,T, C and There is evidence that epigenetic [Pg.158]

Although modifications to DNA and histones are eommonly referred to as being epigenetic, in many cases there is little or no evidenee that they are involved in the epigenetic transfer of information, though they may be involved in the regulation of transeription/expression in individual eells or organisms. [Pg.159]

In the following sections, we outline the features of the enzymes responsible for catalysing the placement and removal of post-translational modifications of histones, with particular focus on structure, mechanism and the potential for pharmacologically relevant inhibition. We also examine some of the proteins that recognise these modifications by selectively binding to them. We discuss, where applicable, the biological roles of some of these proteins, and the links to human disease. [Pg.159]

Fluorspar deposits ate commonly epigenetic, ie, the elements moved from elsewhere into the country rock. For this reason, fluorine mineral deposits ate closely associated with fault 2ones. In the United States, significant fluorspar deposits occur in the Appalachian Mountains and in the mountainous regions of the West, but the only reported commercial production in 1993 was from the faulted carbonate rocks of Illinois. [Pg.171]

Epigenetic carcinogen A substance that causes cancer through a mechanism other than interaction with the genetic material. [Pg.1435]

Many anticancer agents currently in use, including chemotherapeutic drugs and radiation, are potent inducers of apoptosis and cell-cycle arrest. It is believed that induction of these molecular pathways is central to the efficacy of such agents [1]. Genetic and epigenetic alterations that contribute to tumorigenesis... [Pg.317]

In higher eukaiyotes, most of the chromosomal DNA carries 5-methyl-cytidine residues located in CpG sequence motives. There is a close correlation between transcriptional inactivation and methylation. On the other hand, considerable evidence shows that regions of DNA that are actively engaged in transcription lack 5-methyl-cytidine nucleotides in CpG motivs. Hence DNA methylation is a means how cells regulate gene expression. DNA methylation which is catalyzed by DNA methyltransferases is the best characterized epigenetic mechanism. [Pg.432]

An important aspect of the histone code theory is that some modifications can be passed on during cell division. As a result histone modification patterns including histone acetylation serve as a means to store inheritable traits of an organism which are not DNA encoded. This kind of information is generally termed epigenetic information. [Pg.594]

Nasmyth No, that is what we know about these epigenetic states they are either on or they are off. [Pg.112]
