Chronic HCV- In combination with interferon alfa-2b injection for the treatment of chronic HCV in patients 3 years of age (oral solution) or 5 years of age (capsules) and older with compensated liver disease previously untreated with alpha interferon or in patients who have relapsed following alpha interferon therapy. Note Ribaspheres is only indicated in combination with interferon alfa-2b in patients 18 years of age and older. [Pg.1772]

Alpha interferons, including peginterferon alfa-2a, may cause or aggravate fatal or life-threatening neuropsychiatric, autoimmune, ischemic, and infectious disorders. [Pg.1980]

Autoimmune disorders Development or exacerbation of autoimmune disorders, including myositis, hepatitis, idiopathic thrombocytopenic purpura, psoriasis, rheumatoid arthritis, interstitial nephritis, thyroiditis, and systemic lupus erythematosus have been reported in patients receiving alpha interferon. [Pg.1989]

Bone marrow toxicity Peginterferon alfa-2a suppresses bone marrow function and may result in severe cytopenias. Ribavirin may potentiate the neutropenia and lymphopenia induced by alpha interferons including peginterferon alfa-2a. Alpha interferons may be associated with aplastic anemia very rarely. [Pg.1989]

Colitis Ulcerative and hemorrhagic/ischemic colitis, sometimes fatal, has been observed within 12 weeks of starting alpha interferon treatment. Abdominal pain, bloody diarrhea, and fever are the typical manifestations of colitis. [Pg.1989]

Hypersensitivity reactions Severe acute hypersensitivity reactions (eg, urticaria, angioedema, bronchoconstriction, anaphylaxis) have been rarely observed during alpha interferon and ribavirin therapy. [Pg.1990]

Ophthaimoiogic disorders Decrease or loss of vision, retinopathy (including macular edema, retinal artery or vein thrombosis), retinal hemorrhages and cotton wool spots, optic neuritis, and papilledema are induced or aggravated by treatment with peginterferon alfa-2a or other alpha interferons. All patients should receive an eye examination at baseline. [Pg.1990]

Pancreatitis Pancreatitis, sometimes fatal, has occurred during alpha interferon and ribavirin treatment. [Pg.1990]

Puimonary disorders Dyspnea, pulmonary infiltrates, pneumonia, bronchiolitis obliterans, interstitial pneumonitis, and sarcoidosis, some resulting in respiratory failure and/or patient deaths, may be induced or aggravated by peginterferon alfa-2a or alpha interferon therapy. [Pg.1990]

Alpha interferons, including peginterferon alfa-2b, cause or aggravate fatal or life-threatening neuropsychiatric, autoimmune, ischemic, and infectious disorders. Closely monitor patients with periodic clinical and laboratory evaluations. Withdraw from therapy patients with persistently severe or worsening signs or symptoms of... [Pg.1992]

Neuropsychiatric events Life-threatening or fatal neuropsychiatric events, including suicide, suicidal and homicidal ideation, depression, relapse of drug addiction/overdose, and aggressive behavior have occurred in patients with and without a previous psychiatric disorder during peginterferon alfa-2b treatment and follow-up. Psychoses, hallucinations, bipolar disorders, and mania have been observed in patients treated with alpha interferons. [Pg.1998]

Pancreatitis Fatal and nonfatal pancreatitis have been observed in patients treated with alpha interferons. [Pg.1999]

Elderly Treatment with alpha interferons, including peginterferon alfa-2b, is associated with CNS, cardiac, and systemic (flu-like) adverse effects. Because these adverse reactions may be more severe in the elderly, exercise caution in the use of interferon alfa-2b in this population. [Pg.2000]

The DNA-mediated effects of fluropyrimidines can be modulated by a number of agents, such as leucovorin (LV), levamisole, and interferon-alpha (IFN-alpha). LV prolongs TS inhibition by increasing the availability of the reduced folate cofactor necessary for formation of the inactive TS-FdUMP complex (21) (Fig. 4). Studies show alpha-interferon can potentiate 5-FU-mediated cyotoxicity, but the mechanisms are not yet defined (22,23). Another approach to modulate the activity of fluoropyrimidines is the use of the nucleoside transport inhibitor dipyridamole. Dipyridamole probably permits... [Pg.27]

A type of interferon produced by white blood cells, alpha interferon, has been engineered and produced to keep the progression of hepatitis C virus, a serious liver infection, under control. The first alpha interferons were slightly modified forms of natural interferon alpha produced in E. coli. More recent versions, peginterferon and... [Pg.75]
