In order to prepare C -labeled d-a-tocopherol we started from natural d-7-tocopherol (Fig. 6) and introduced the C -methyl group into the 5-position by use of the Mannich reaction. Condensation of d-y-tocopherol with radioactive paraformaldehyde in the presence of diethylamine followed by catalytic reduction gave d-a-tocopherol. This was acetylated to methyl-C Mabeled d-a-tocopherol acetate (specific activity 1.7 tc/mg Fig. 6). [Pg.395]

The a-tocopherol, P-carotene (ATBC) Cancer Prevention study was a randomised-controlled trial that tested the effects of daily doses of either 50 mg (50 lU) vitamin E (all-racemic a-tocopherol acetate), or 20 mg of P-carotene, or both with that of a placebo, in a population of more than 29,000 male smokers for 5-8 years. No reduction in lung cancer or major coronary events was observed with any of the treatments. What was more startling was the unexpected increases in risk of death from lung cancer and ischemic heart disease with P-carotene supplementation (ATBC Cancer Prevention Study Group, 1994). Increases in the risk of both lung cancer and cardiovascular disease mortality were also observed in the P-carotene and Retinol Efficacy Trial (CARET), which tested the effects of combined treatment with 30 mg/d P-carotene and retinyl pahnitate (25,000 lU/d) in 18,000 men and women with a history of cigarette smoking or occupational exposure to asbestos (Hennekens et al, 1996). [Pg.33]

Procedure Transfer an accurately measured volume of the Test Solution, equivalent to about 100 mg of a-tocopherol, into a separator, and add 200 mL of water. The a-tocopherol molecule has three centres where stereoisomers can occur. The naturally occurring molecule is the D-a-tocopherol (or RRR-a-tocopherol) configuration and has the highest vitamin activity. Synthetic DL-a-tocopherol acetate (also called aU racemic a-tocopherol acetate) is used as a vitamin E supplement and comprises aU eight possible stereoisomers only one molecule in eight is in the RRR form. The vitamin activity of the four stereoisomers in the L forms is considerably lower than the four that make up the D forms in the latter the RRR form is the most active. [Pg.82]

The d-form of the vitamin is the most active. Any studies of vitamin supplementation, and their subsequent effects in various muscle foods, must acknowledge the form of tocopherol fed before conclusions can be drawn regarding dietary uptake and antioxidant efficiency, d-a-tocopherol or its ester is more readily assimilated into tissues than the racemic (dl) form (Hidiroglou et al., 1988). Burton et al. (1988) have demonstrated that the uptake of the free phenol and acetate forms of d-a-tocopherol are equivalent. Marusich et al. (1975) reported that dl-a-tocopherol and dl-a-tocopheryl acetate were equivalent in terms of uptake by chicken liver and breast muscle, and resulted in similar oxidative stabilities within the tissue types. [Pg.162]
