The mitotic index is the fraction or percentage of cells in mitosis within a given cell population. The thymidine labeling index is the fraction of cells incorporating radioactive thymidine. They represent cells in M-phase and S-phase and define the proliferative characteristics of normal and tumor cells. [Pg.630]

The results of incorporation experiments using tritiated thymidine lend support to the hypothesis that thymidine rhamnosyl pyrophosphate in Lactobacillus acidophilus is an intermediate on the pathway to deoxyribonucleic acid synthesis. When cells were grown in a medium containing tritiated thymidine, labeled thymidine rhamnosyl pyrophosphate constituted two-thirds of the total label in the acid-soluble fraction. When these cells were transferred to a medium containing 2-deoxyadenosine, 2-deoxy-guanosine, and 2-deoxycytidine as the nucleosides, 99 % of the label initially present was transferred to deoxyribonucleic acid after incubation for 50 minutes. The adsorption and penetration of AAV containing H-thymidine-labeled DNA has been measured with KB cells in spinner culture (Rose and Koczox, 1972 Berns and Adler, unpublished data). Adsorption reached a maximum and plateaued one to two hours after infection. Approximately 16-30% of the added viral label was irreversibly associated with the cells by this time. Depending on the method of cell fractionation, 25-45% of the bound radioactivity was eventually (by 20 hours) found in the nuclear fraction, the majority within two hours. Uncoating, as measured by sensitivity of the labeled DNA to DNase, took place in the nucleus.
