Mitotic index

Hryant I am a little concerned about going straight from mitotic index to proliferation rate. Is that a valid assumption to make for these cells Really, the mitotic index only tells you what fraction of the cells are in mitosis, and it could be the same for a fast-growing cell population as a slow one.

Raff Also, if there are more stem cells, and these are dividing more slowly, this alone could explain the decrease in the mitotic index in the mutant.

At 24 hours, a strong decrease in the number of topi-positive cells and in topi staining intensity appeared in tumors treated with irinotecan, whereas there was almost complete disappearance of topi staining in tumors treated with edotecarin (Figure 11D). Morphologic analysis of treated samples show absence of mitotic figures after both treatments (mitotic index 2% in controls, 0% in treated samples data not shown). 

The mitotic index is the fraction or percentage of cells in mitosis within a given cell population. The thymidine labeling index is the fraction of cells incorporating radioactive thymidine. They represent cells in M-phase and S-phase and define the proliferative characteristics of normal and tumor cells.

Because the location of mitosis is fixed and its duration short, one can, however, say that the proportion of cells in mitosis, the mitotic index,...

In order to produce a stirring rate of 150 r.p.m. Crespi and Thilly (1982) made a special harvesting vessel with stirring bars. Their selected cells had a viability of 80-95% and a mitotic index of 85-95%. 

Process the cells for autoradiography ( 12.3) and score the percent of cells labelled and the mitotic index. 

Readdition of isoleucine leads to induction of DNA synthesis in most cells starting at 4-5 h and the mitotic index begins to increase at 12 h in CHO cells.
