This technique can be applied to samples prepared for study by scanning electron microscopy (SEM). When subject to impact by electrons, atoms emit characteristic X-ray line spectra, which are almost completely independent of the physical or chemical state of the specimen (Reed, 1973). To analyse samples, they are prepared as required for SEM, that is they are mounted on an appropriate holder, sputter coated to provide an electrically conductive surface, generally using gold, and then examined under high vacuum. The electron beam is focussed to impinge upon a selected spot on the surface of the specimen and the resulting X-ray spectrum is analysed. 

A section of the film was removed from the bag and was immersed in liquid nitrogen. The section was then freeze-fractured and mounted on the SEM stage with the outside surface of the bag section facing downward. The sample was sputter coated prior to SEM/EDS analysis. Note We recommend that after performing step 1 above that the specimens be rinsed three times in a phosphate buffer (pH 7.2), which will remove any residual glutaraldehyde before beginning the dehydration series. Garduno etal. (13) also use a second technique in which they cut small pieces of medium containing diatoms and immediately fix the pieces in liquid nitrogen. The pieces are freeze-dried and then gold sputter coated.

A cross-sectional view of a C. digitata seed is shown in a scanning electron micrograph (SEMT in Figure 1. A scanning electron microscope, SEM (JEOL JSM-5300, JEOL USA, Inc., Peabody MA) was used to examine the morphology of dentin surfaces treated with solutions of various PIDAA derivatives at concentrations of 0.1 and 0.3 mol/L. Levels of lactate in buttermilk and yoghurt (and blood) were estimated using disposable sensors formed from screen-printed graphite laminated between two polymer sheets [18]. Microscopy. Scanning electron microscopy was run on resin samples using a AMR 1200 Scanning Electron Microscope. The samples were mounted on the stub using double stick tape and then sputter coated with gold. 

Hair fibers are mounted with double-stick tape on aluminum stubs, sputter coated with a 4 nm layer of gold, and examined at 20 kV at a working distance of approximately 15 mm in our hands on a Hitachi S3000N VP Scanning Electron Microscope (83).
