What can be expected from the future The cultivation of isolated hematopoietic cells will benefit from the identification of new cytokines, making the use of stromal-conditioned medium unnecessary. The recent development of proteomics techniques will expediate this process. The prospects of the co-cultivation will depend on the development of human stromal cell lines supporting the expansion of early progenitor cells, and the membrane approach has to prove its potential on a clinical scale. 

ECVAM is the leading international center for alternative test method validation. Hartung et al. (29) summarized the modular steps necessary to accomplish stage 3 (test validation). The seven modular steps are (I) test definition, (2) within-laboratory variability, (3) transferability, (4) between-laboratory variability, (5) predictive capacity, (6) applicability domain, and (7) performance standards (29). Steps 2-4 evaluate the test s reliability steps 5 and 6 evaluate the relevance of the test. Successful completion of all seven steps is necessary to proceed to stage 4 (independent assessment or peer review). This modular approach allows flexibility for the validation process where information on the test method can be gathered either prospectively or retrospectively. The approach is applicable not only to in vitro test methods but also to in silico approaches (e.g., computer-based approaches such as quantitative structure-activity relationships or QSAR) and pattern-based systems (e.g., genomics and proteomics).

Discriminating on one hand the bead type and on the other hand the hybridized sequence leads to a sensitive and high throughput technique with detection limits in the lOfmole range [24]. Given this perspective that proteomics is not so much a new field as the substantial expansion of an old one, it is germane to briefly consider where this synthesis of old and new technologies has brought us in terms of some of the more active areas of research before discussing some of the future prospects. Table 3 lists four such areas, which not surprisingly correlate quite closely to the definitions described in Table 1. There is necessarily some overlap between these.
