The effect of BHA on the In vivo formation of BP metabolite-DNA adducts in the forestomach of female A/HeJ mice was also examined (7) In this study, mice were fed BHA, 0.5% of diet, for eighteen days. H-BP mg/mouse) was adminitered and the animals were sacrificed 48 hours later. As in lung and liver, the BPDEI-deoxyguanosine adduct was the predominant adduct formed in the forestomach. BHA treatment inhibited the formation of BPDE-DNA adducts in the forestomach by 50%. Wattenberg (8) has shown that BHA treatment inhibits BP-induced forestomach tumors approximately 90%. Although our protocol for administration of BP was not identical to that used in the tumorigenicity studies, we can still see a definite trend between BHA inhibition of BPDE-DNA adduct formation in the forestomach of A/HeJ mice and reduction in tumor formation. [Pg.244]
