Analysis suspended matter

The PEG, MCPEG and DCPEG metabolites were monitored in marine, estuarine, river and wastewater near and in the Italian river Tiber. Water samples were analysed by LC—ES—MS analysis. Suspended particulate matter (SPM) was also analysed, but none of the metabolites was detected in this matrix. 

Pharmacopoeial methods rely heavily on simple analysis by UV/visible spectrophotometry to determine active ingredients in formulations. These methods are usually based on the use of a standard A (1 %, 1 cm) value for the active ingredient being assayed and this relies on the UV spectrophotometer being accurately calibrated as described earlier in the chapter. Such methods also presume that there is no interference from excipients (preservatives, colourants, etc.) present in formulations and that the sample is free of suspended matter, which would cause light scattering. 

The instruments discussed earlier are the primary ones used in toxicant analysis, but an enormous number of analytical techniques are used in the field. Many of the instruments are expensive (e.g., Raman spectrometers, X-ray emission spectrometers) and few laboratories possess them. Many other instruments are available, however, such as the specific-ion electrode, which is both sensitive and portable. Specific-ion electrodes have many other advantages in that sample color, suspended matter, turbidity, and viscosity do not interfere with analysis therefore many of the sample preparation steps are not required. In applying the PEEP index concept to sets of industrial effluents thus far, wastewater samples have been filtered prior to bio-analysis (see Section 5.1). Hence, only their soluble toxicity potential is taken into consideration. This is certainly a drawback at this time as toxic and genotoxic potential linked to suspended matter of some industrial plant effluents, for example, have been shown to be important (White et ah, 1996 Pardos and Blaise, 1999). Particulate toxicity in effluent samples should certainly be addressed in future PEEP applications, as soon as reliable small-scale toxicity tests are developed and available to estimate it. Indeed, the issue of soluble and particulate toxicity is especially relevant in relation to technology-based reduction of hazardous liquid emissions.

For phytoplankton particulate organic carbon (POC) and particulate nitrogen (PON) determination, some suspended matter was collected on precombusted (450°C) Whatman GF/F filters, dried at 60°C and stored in polystyrene Petri dishes until analysis. The samples were first filtered through 0.45 pm Millipore HA filters to remove suspended matter and micro-organisms, and then distributed into conditioned polyethylene bottles where they were kept at -18 until analysis. 

Sampling and measurement of this class requires dynamic sampling equipment since this class of air pollutants represents particles and droplets too small to fall out of their own accord. Work has to be done on the gas to force it through the recovery or analytical equipment to capture the suspended matter. The analysis can be a source test for which a stack or waste vent is sampled directly, or it can be an ambient air survey, where the general condition of the outside air is determined.

Lack of homogeneity of the sample may also be a source of deviation from the Lambert—Beer law in view of the increased possibility of radiation losses at the liquid/liquid interfaces, as well as the presence of gas and/or liquid mini-bubbles and suspended matter (see also Fig. 4.1). It was soon realised that DWS was also useful for the analysis of turbid samples. Analogously to spectrophotometry, stray radiation can alter the measured turbidance (Eq. 4.6), the limitation becoming more severe at lower transmitted power. The presence of suspended matter in the processed sample is inherent to turbidimetry and leads to an amplification of stray light due to scattering effects. This effect has not been systematically investigated in flow analysis.

The quantity of isolated suspended matter should be exactly determined and the concentration of suspended particles is used as a characterization of the investigated water basin. The concentration of metals in the filtered water is actually only the quantity of dissolved metals therefore separate analysis of the suspended matter may be recommended for evaluation of the total content. 

The techniques are suitable for the analysis of estuarine waters whose high and variable salinity and suspended matter content creates considerable diflBculties in analytical techniques designed for use in ocean waters and fresh waters. The precision and accuracy of the developed techniques has been determined under the entire range of conditions encountered in San Francisco Bay. Several methods of sample preservation have been investigated and methods that are reliable in preserving nitrogen and phosphorus forms in San Francisco Bay water are recommended. 

The sample to be analyzed should first be filtered through a micropore membrane filter of 0.45 micron average pore diameter. This eflFec-tively removes suspended matter and insures subsequent determination of only that material considered to be in true solution. The suspended material may then be examined by x-ray analysis. If the thioacetamide method were used to analyze the unfiltered sample, some of the suspended sediment might not dissolve and would remain to contaminate the sulfide precipitate.
