Conceivably, lessons learned in yeast on internalization of membrane proteins are relevant to both the constitutive and the inducible pathways of receptor endocytosis in mammals. The constitutive pathway is the least understood, and it may bypass a requirement for ubiquitination by utilizing intrinsic di-leucine and other signals for protein internalization (Covers et al. 1998). In the ErbB family, the pathway may be exemplified by the relatively slow internalization rate of mutant RTKs, either the kinase- defective ErbB-3 (Baulida et al. 1996 Baulida and Carpenter 1997 Waterman et al. 1998), or artificial kinase-defective mutants (Chen et al. 1989 Felder et al. 1990).

Prior to the mid-1980s the chemical industry experienced some upheavals of which the thalidomide tragedy of the early 1960s and the Bhopal catastrophe in 1984 are two major examples. However, the lessons learned from such sad events led to a spectacular improvement during the 1990s in the safety record of the industry in the Western world.

The chemical industry generally handles business so well that it is difficult to find large numbers of recent incidents for examples. Many of the featured case histories in this book occuned over 15 years ago however, the lessons that can be learned will be appropnate into the twenty-first century. Tanks can fail from the effects of overpressure and underpressure in the 1990s just as easily as they failed in the 1970s. Incompatible chemicals are incompatible in any decade and humans can be forgetful at any time. Before we review a single ca.se history, it is time to boast about the chemical industry s safety record.
