DETA/NO is a stable NO-donor with the longest NO generation half-life of approximately 20 h. Thrombelastography performed on rabbit blood showed that DETA NONOate-derived NO significantly decreased coagulation activity and platelet activation [48]. Monitoring by intravital microscopy showed that DETA/NO attenuated the platelets/endothelial cells adhesion response to endotoxins (e.g. lipopolysaccharides) in murine intestinal venules [49]. The main mechanism of the antiadhesive action of DETA/NO on platelets was activation of soluble guanylate cyclase [49].

VE)-cadherin and platelet endothelial cell adhesion molecule-1 (PECAM-1). VE-cadherin is an endothelial-specific Ca " -regulated protein that is linked to the cytoskeleton via catenins (Fig. 1). PECAM-1, also known as CD31, is a key participant in the migration of blood-borne cells across the BBB. Changes in the adherent junction proteins can lead to increased paracellular permeability (Abbruscato and Davis, 1999) and leukocyte trafficking in the CNS (Newman, 1994 Garrido-Urbani et al., 2008).

Other members of the Ig-superfamily function in conjunction with counter-receptors from separate families of adhesion molecules in various events such as leukocyte trafficking and the movement of immune cells and probably tumor cells across cell barriers in inflammation and metastasis. These glycoproteins (Fig. 10) are the immune cell adhesion molecules ICAM-1, -2 and -3, the vascular cell adhesion molecule VCAM and the platelet-endothelial cell adhesion molecule PECAM [131-134].
