In human B-cell lines, activators like phorbol esters (PMA) and bacterial lipopoly-saccharide (LPS) upregulatethe concentrative transporters, whereas the equilibrative transporter hENTl is downregulated. This effect can also be produced by TNF-a,... [Pg.58]

Epstein-Barr virus (EBV)-transformed homozygous human B-cell lines were used as a source for isolating HLA class II molecules. In the case of isolation of HLA-DR1, WT-100 cell pellets were lyzed by NP-40, and DR1 was isolated from homogenates by affinity chromatography with the monoclonal antibody L243 essentially as described [64,65].The purity of the preparation was analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), HPSEC and Western blotting. [Pg.363]

Preparation of cell extracts. MOLT 4F is a human T-cell line established from a patient with acute lymphocytic leukemia (13) and has since been maintained in our laboratory. Characteristically, MOLT 4F cells form rosettes with sheep red blood cells (E-rosettes). They do not synthesize immunoglobulin and grow as a single cell suspension. RPMI 1788 is a human B-cell line established from the peripheral blood of a healthy volunteer. These cells produce immunoglobulin, have complement receptors and grow as clusters in suspension culture. Characteristics of these cell lines have recently been summarized by Moore (14). These cells were maintained in medium RPMI 1640 with 10% fetal calf serum and penicillin and streptomycin. The viability of the cells measured by a trypan blue... [Pg.154]

In reversal experiments using the human B-cell line (RPMI 8422), pyrazofurin inhibited cell growth at a concentration of 10 M. This PF effect was completely reversed with the addition of 10 M to 10 M of uridine or cytidine but not by orotic acid or orotidine. This is additional confirmation of the assumption that the site of inhibition in the biosynthesis of pyrimidine nucleotides is at the site of orotidylic carboxylase ... [Pg.160]

Two new polycyclic pyridoacridine alkaloids, arnoamine B, 289, and its demethylated analogue, arnoamine A, have been isolated from the ascidian Cystodytes sp. These new compounds are the first known examples of pyrroles fused to pyridoacridines, have antifungal properties, and are cytotoxic in several human tumor cell lines <1998JOC1657>. Both these compounds have been synthesized, starting from the hydrazone 290 with a Fischer indole synthesis (Scheme 73) <2000JOC5476>. [Pg.915]

Fusion of human lymphocytes with human lymphoblastoid cell lines is a very inefficient process. Fusion of human lymphocytes with murine myeloma cells lead to very unstable hybrids. Upon fusion, preferential loss of human genetic elements is often observed. Unfortunately, particularly common is the loss of chromosomes 2,14 and 22, which encode antibody light and heavy chain loci. The production yields of human monoclonals upon immortalization of the human B-lymphocyte (by whatever means) are also low. [Pg.392]

Mature B lymphocytes express AhR and ARNT,10,12 and the ability of TCDD to suppress primary antibody responses has been known for a long time (reviewed in4). While the precise mechanisms resulting in suppression of T-dependent B cell responses in vivo have not been fully elucidated, it is clear that direct effects of TCDD on B cells plays a role. Studies using two B cell lines with differential AhR expression have linked suppression of LPS-induced IgM production to AhR activation, and have shown that AhR interactions within the 3 alpha immunoglobulin heavy chain enhancer directly influence antibody production.52-54 TCDD also appears to directly modulate the expression of CD19 on human B cells, suggesting another mechanism by which TCDD down-modulates the responsiveness of mature B cells.55... [Pg.243]
