Fast-dissolving formulations (flash-dispersing) are not primarily intended to be for buccal delivery the issue here is that they may be taken without water. This causes an important difference in performance relative to ordinary immediate-release products, especially if the drug is in suspension. If the material is swallowed dry, it may adhere to the fundus area, where the amount of shear is low. This causes a significant fraction of the material to be retained resulting in tailing of the absorption phase and an apparently decreased AUC as the material is released over several hours. [Pg.550]

Agricultural Products. Pesticides are frequently applied as emulsifiable concentrates. The active insecticide or herbicide is dissolved in a hydrocarbon solvent which also contains an emulsifier. Hydrocarbon solvent selection is critical for this application. It can seriously impact the efficacy of the formulation. The solvent should have adequate solvency for the pesticide, promote good dispersion when diluted with water, and have a flash point high enough to rniriinii2e flammability hazards. When used in herbicide formulas, low solvent phytotoxicity is important to avoid crop damage. In contrast, there are no solvents used in HME processes for manufacturing amorphous solid dispersions. Instead, the mixture of crystalline drug and polymer are heated to a temperature at which the components melt or form a eutectic, and then flash cooled, resulting in a dense amorphous glassy solid. The solid is then milled to achieve uniform particle size distribution, which is then processed into the final formulation.
