Duclauxin (9) was identified (14) as one of the agents responsible for the antitumor activity of culture filtrates of P. stipitatum. It was shown that the phenalenone decreased the proliferation of cells of Ehrlich s ascites carcinoma in vitro, and that the compound exerts its cytotoxic effect on cancer cells by inhibiting the synthesis of nucleic acids without affecting protein synthesis (40).

The in vivo distribution of a new, polymer-nitroxide probe, spin-labelled dextran (termed TEMPO-DX) was investigated by Saito et al, who also compared its distribution and pharmacokinetics to that of 3-carbamoyl-proxyl. As expected, it was found that the half-life of the larger molecule was considerably longer than that of 3-carbamoyl-proxyl, and that the TEMPO-DX concentrated in the tumour of a mouse with an Ehrlich s ascites carcinoma.

Ehrlich ascites carcinoma cells implanted intra-peritoneally in female mice were reduced in their viability and volume by oral supplementation of melatonin at 50mg/kg body weight (El-Missiry and El-Aziz 2000). Flow cytometric studies showed that melatonin not only delayed the progression of cells from GO/Gl phase to S-phase of the cell cycle but also reduced DNA synthesis during cell cycle. In addition, the aneuploidy status was depressed in melatonin treated mice.

Emetine - As an inhibitor of protein synthesis at the transcription level, emetine is also active against leukemias L-1210 and P-388, B16 melanomas, Ehrlich ascites carcinoma and Yoshida sarcoma. Pharmacologic studies showed that it effects a sympathetic blockade, antagonizes hyal-uronldase, inhibits oxydative N-demethylatlon of aminopyrine and N-ethyl-morphlne as well as the S-demethylation of 6-MP riboside in vitro. 2,143... 

Diphtheria toxin, a protein secreted by lysogenic strains of Corynebacterlum dlphtherlae, has been shown to be an effective inhibitor of the growth of Ehrlich carcinoma, both ascitic and solid, in mice. The effect is thou to be mediated primarily through inhibition of protein synthesis. Similarly, the parasporal crystal ( S -endotoxin) of B. thuringiansis has been reported to exertj tent anti-Yoshlda ascites carcinoma activity, with a hi therapeutic index . 

Heparin causes a reduction in mitotic index and a decrease in tumour volume for Ehrlich ascites tumours in mice . Polyethylene sulphonates having molecular weights between 15,000 and 35,000, also have antineo-plastic activity in mice against ascites tumours, carcinomas and leukaemias . Heparin has been found to reduce markedly the number of pulmonary metastases in rats receiving intravenous Walker 256 carcinoma cells . Evans blue in the presence of a sulphated pectin entered tumour cells but not normal tissue cells s. The anti-immunochemical action of heparin has been used successfully for tumour heterografts Ass. 

Earlier, some of the same considerations had led me in 1957 to initiate a study of amphibian venoms of the steroidal bufadienolide (6) type as potential sources of new antineoplastic substances. Eventually we found that some of the toad venom bufadienolides such as marinobufagin significantly inhibit growth of the National Cancer Institute s KB cell line derived from a human nasopharynx carcinoma and lead to a curative response with the murine Ehrlich ascites system (7). However, the therapeutic indices were unattractive for further development. So the effort was extended in 1965-66, as Just noted, to encompass a geographically far-reaching area (Asia, Africa, Australia etc.) and an extensive research program to evaluate marine invertebrates and arthropods for structurally unique and useful anticancer constituents. Subsequently we isolated the first such invertebrate antineoplastic constituents (8-12). Meanwhile, our early expectations have been abundantly realized and the discovery of the bryostatins provides a splendid illustration.
