Like plasma and urine, matrixes from plant or environmental sources contain a vast diversity of components. Thus, achiral-chiral LC-LC is also useful for analysis involving samples from these sources. Stalcup et al. (1991) studied the enantiomeric purity of scopolamine extracted from Datura sanguinea in both homogenized plant leaves and commercial extracts. The 23-cm-diameter distillation column under study is used to separate ethanol and water. The separation of chemical species of elements can only be performed by techniques which do not destroy the chemical forms, e.g. possible degradation of the species upon heating of the column. A direct injection nebuliser (DIN) was used to interface LC with ICP-MS (Shum et al., 1992a). The DIN transferred all of the sample into the inductively coupled plasma. Microscale LC separations in small packed columns were studied because the column flow rates of about 30 ml min 1 were compatible with the DIN. The low dead volume (less than 1 ml) of the interface prevented excessive band broadening. Eluents containing up to 85% methanol were accommodated. The analyte signal varied by about 20% as the eluent changed from 20% to 80% methanol in water. The effect of temperature, and of temperature and pH on the retention of a selected group of compounds using a beta-cyclodextrin column was studied. In a separate study, the quantity of thalidasine in a polyphase-liposome injection (139-2) was determined by dilution of the injection with EtOH-O.OlN HC1 (4 1), followed by spectrofluorometric determination at 235 nm (for excitation) and at 318 nm (for emission).
