An alternative method for assessing cell layer integrity is through the use of hydrophilic paracellular transport markers (e.g., radiolabeled D-mannitol or fluorescein-Na+), which passively traverse cells by the paracellular route. Small amounts of compound required for in vitro conjunctival cell culture transport experiments make this approach well suited for screening purposes. Relative absorption index of a series of pharmacologically active molecules can be ranked against known markers for the identification of candidates with potential absorption problems, which is a reliable tool to select drug candidates with optimal characteristics. [Pg.317]

Level 3 of the screen is designed to determine the cytotoxic selectivity of samples for tumor cells vs. normal cells. Where possible, the same patient s tissues are used. As in Level 2, six serial dilutions (of four- to five-fold each) are assayed in triplicate for each sample. The diluted samples are then added to the tumor cell and normal cell cultures, and the IC50 is determined. A "selectivity index" (SI) is determined based on the IC50 for normal cells/IC5Q for tumor cells. Samples with an SI of three or more are advanced to Level 4 of the screen. The infected cell culture is brought into 50-mL tubes and the cells are pelleted, and the supernatant is carefully distributed in aliquots of 1 mL in cryotubes. The cryotubes are labeled with a virus stock number. The number and the content are indexed on the list of virus stocks and the tubes are stored at -80°C. For delicate strains, the freezing procedure might damage the virus. Therefore, the titration of the virus stock should be done on a similar frozen aliquot as for the susceptibility testing.
