Both TPA and PMA enhance carcinogenesis by binding to the DAG binding site and activating PKC, which bypasses normai ceii cycie reguiation and stimulates cell division to produce its "tumor promoting" effect. 

Tumor-promoting effect. A 28-year-old man who abused alcohol, nicotine, and cannabis for several years was investigated. He suffered simultaneously from a squamous cell carcinoma of the hypopharynx with bilateral cervical metastases, an adenocarcinoma of the transverse colon and a primary hepatocellular carcinoma. There were occurrences of three separate malignant tumors with different histologies in the aerodigestive tract, which could be related to a chronic abuse of cannabis . 

Toxic effect. Water extract of the dried leaf, administered orally to adults, was inactive " ". Infusion of the dried leaf, administered in drinking water to male rats at a dose of 5%, was inactive "". Tumor-promoting effect. The tumorigenic effect of benzo[a]pyrene was retarded by the creosote oil, neutral distillate, neutral residue, and phenolic fractions. However, in three instances this effect was apparently related to skin damage associated with the treatments rather to a specific inhibitory effect. Only the phenolic fraction seemed to exhibit a primary antagonistic effect. Three other fractions (basic, neutral, and a low concentration of neutral distillate) exhibited apparent tumor promoting effects.

Skisak (1991) tested the tumor-promoting activity of MD API 81-07, a hydrosulfurized kerosene, in CD-I mouse skin treated with DMBA. MD API 81-07 was applied 2 times/wk for 25 wk to the skin of treated mice. MD API 81-07 had tumor-promoting effects, inducing squamous cell carcinoma and papilloma of the skin. As already shown, tubeimoside 1 (5) from Bobolstemma paniculatum exhibited a strong anti-tumor activity, but had intriguingly a potent antitumor-promoting effect in two-stage mouse skin tumor formation induced by DMBA+TPA [38]. Its natural analog, tubeimoside III showed also a potent anti-tumor-promoting effect in the same model after topical administration but not after oral administration [39].

Weak tumor-promoting activity was seen in mice treated topically once a week for 33 weeks with eucalyptus oil after application of the carcinogen 9,10-dimethyl-l,2-benzanthra-cene (DMBA). The tumor-promoting effect was less than that of orange or lemon essential oils (Roe and Field 1965). 

A number of studies using cyclooxygenase inhibitors provided strong evidence that the tumor-promoting effects of a high linoleic acid diet are mediated in part through eico-sanoids, particularly the cyclooxygenase pathway of arachi-donic acid (50-52).
