In January 2008, Shim and Wanasundara announced their development of a sandwich ELISA that targets the Sin a 1 allergen from S. alba [11]. The assay was developed to quantify Sin a 1 levels in various yellow mustard seed lines, but was reported to be less sensitive than the previously described B.juncea assay developed by Koppleman et al. Sesamin has been associated with the reduction of serum and liver lipids (Hirose etal., 1991,1996 Ide etal., 2001 Yasumoto etal., 2001). Hirose etal. (1991) reported that sesamin decreased lymphatic absorption of cholesterol. Sesamin was found to be a promoter of hepatic fatty acid oxidation in rats (Ashakumary et al., 1999). Addition of a 1 1 mixture of sesamin and episesamin to the diet of rats (0.1-0.5% of total diet) promoted mitochondrial and peroxisomal palmitoyl coenzyme A (CoA) oxidation rates in a dose-dependent manner. Rats fed diets containing 0.5% sesamin had 2 fold higher mitochondrial activity and 10 fold higher peroxisomal activity than rats fed non-sesamin diets. Yasumoto et al. (2001) reported similar findings when a 20% sesame seed diet was fed to rats. These authors compared two experimental sesame seed lines (0730 and 0732) with the commercial cultivar Masekin. Both experimental seed lines enhanced mitochondrial and peroxisomal fatty acid oxidation rates, and were more effective than the commercial cultivar at reducing plasma triacylglycerol levels (Yasumoto et al., 2001). A study of the effects of dietary sesamin on the hepatic metabolism of arachidonic and eicosapentaenoic acids in rats showed similar increases in hepatic mitochondrial and peroxisomal fatty acid oxidation (Sawada et al., 2001). [Pg.86]

The separation of the solid phase does not occur readily with some liquid mixtures and supercooling is observed. Instead of an arrest in the cooling curve at /, the cooling continues along a continuation of c/ and then rises suddenly to meet the line f g which it subsequently follows (Fig. 1,13, 1, iii). The correct freezing point may be obtained by extrapolation of the two parts of the curve (as shown by the dotted line). The Berlin City electrical engineer M. Kallmann reported in 1899 on a system for controlling stray currents of electric railways [64]. Tissue Culture Assay. Kogure et al. (48) report a novel tissue culture assay for detecting several types of sodium channel blockers. The mouse neuroblastoma cell line ATCC CCL 131 is grown in RPMI 1640 supplemented with 13.5% fetal bovine serum and 100 pg/ml gentamycin, in an atmosphere of 5% C0 95% air at 37 C. Ninety-six well plates are seeded with 1 x 10 cells in 200 pi of medium containing 1 mM ouabain and 0.075 mM veratridine. Veratridine and ouabain cause neuroblastoma cells to round-up and die. In the presence of sodium channel blockers (e.g., TTXs or STXs), the lethal action of veratridine is obviated and cells retain normal morphology and viability. An important feature of this assay is that a positive test for sodium channel blockers results in normal cell viability. Since bacterial extracts can contain cytotoxic components, this assay offers an advantage over tests that use cell death as an endpoint. The minimum detectable level of TTX is approximately 3 nM, or approximately 1/1000 mouse unit. [Pg.81]

Many in vitro studies have been undertaken to explore the relative effectiveness of isothiocyanates, and associated compounds, as inhibitors of cell growth and inducers of apoptosis in cell lines, and some of these are summarised in Table 4.2.
