In a tiny fraction of cases, a quick formula can be used. For most cases, the analysis uses an options tree, with one leaf per possible outcome. However, this falls prey to the curse of dimensionality —the number of leaves on the tree grows exponentially in the number of risk and decision dimensions considered. Thus only a limited, simple set of situations can be optimized in this way because one has to severely limit the decisions and risks that are considered. Tools available to help automate and simplify options analysis, widely used in pharmaceutical project evaluation, include Excel addons such as R1SK [11] and more graphically based solutions such as DPL [12]. Both of these support the creation and evaluation of decision trees and of influence diagrams Figure 11.2 shows a simple example of each of these. A primer in applied decision theory is Clemen s book Making Hard Decisions, other sources may be found in the website of James Vornov, Director of Clinical Research at Guildford Pharmaceuticals, a recent convert to decision theory for options analysis [13]. [Pg.254]

The newest R D cost study by DiMasi et al. includes seven biotech compounds, in a sample of 69 entities, for which dafa were obfained from major pharmaceutical and bio-pharmaceutical firms. While fhis sample of biological entities is too small to be representative of all biotech development, the clinical phase costs in the DiMasi et al. study were similar for the biotech and pharmaceutical projects. [Pg.539]

An appropriate design team will demonstrate expertise in contamination control philosophies, space planning, code compliance, and mechanical and electrical design and will be familiar with materials of construction currently being used in pharmaceutical projects. It is frequently helpful to include a member of the construction team in the front-end planning effort to advise on constructibility of the facility being planned. Unrealistic construction schedules will be avoided and field rework will be minimized if appropriate attention is paid to the construction phase early in the planning process. [Pg.204]

Recognizing the step-by-step process involved in even the smallest pharmaceutical project can help focus attention in a manner that will result in a successful project. The formal schedule of a well-conceived project will include needs assessment, front-end planning, and preliminary design. It is important that project progress is measured against such a schedule and not just by the visual impact caused by bricks and mortar being installed. [Pg.207]

The first step in a structured approach to the validation of a database system is the Computer System Definition. This is one of the most important steps for any pharmaceutical project, particularly so for a blood establishment database system. Critical considerations include ... [Pg.927]

The experimental determination of protein structures is a difficult task and is not routinely possible. Especially membrane-bound proteins are difficult to crystallize [16]. Therefore, the situation that the target protein structure is unknown is common at an early stage of a pharmaceutical project. In such cases, only the structure of molecules already known to bind to the target protein, for example an endogenous ligand, can be used for virtual screening. [Pg.576]

If the project has sizable architectural considerations, it is important they be properly estimated. Those dollars per square foot numbers that are so useful early in the project need to be firmed up. For the biotech and dosage pharmaceutical projects of today, it is as important that the building costs be as accurate as the equipment costs. Sometimes an account-by-account evaluation for estimating accuracy is in order to see if sufficient work has been done to assure the validity of the overall estimates accuracy. [Pg.765]

Anna-Lena Ungell, PhD, is associate director of DMPK and bioanalytical chemistry at AstraZeneca R D Molndal. She supports preclinical and pharmaceutical projects relating to absorption as well as the gastrointestinal absorption of drugs. Her experience includes absorption models, mechanisms of drug absorption from the intestinal tract, substance evaluation and basic formulation development. Dr. Ungell has four patents and has written numerous research articles and reviews. [Pg.608]

One of the best ways for the computational chemist to influence the drug discovery process is to supply essential information and good ideas, which, when implemented, help drive a pharmaceutical project toward a successful conclusion. In this volume of Reviews of Computational Chemistry, we provide chapters that advance our understanding of the important tools of computational chemistry applicable to computer-aided ligand design. [Pg.442]
