In addition to genotoxicify studies with chlorophenols themselves, the corresponding activity of some of their metabolites has also been examined. The major metabolite of pentachlorophenol in mice and rats, tetrachloro-7 ora-hydroquinone, induced mutations in the hprt locus (but not the ouabain-resistance locus) of V79 Chinese hamster cells (Jansson Jansson, 1991), covalent damage (including 8-hydroxyguanine) in naked DNA in the presence of Cu(Il) (Naito et al., 1994), DNA strand breaks and accumulation of p53 in NIH 3T3 cells in vitro and transfonnation of mouse embryo fibroblasts in a two-stage model (Wang et al., 1997). The same metabolite, as well as tetrachloro-1,4-benzoquinone and tetrachloro-l,2-benzoquinone, caused DNA strand breaks in V79 Chinese hamster cells (Dahlhaus et al., 1996). [Pg.804]

Figure 7-32 Micrograph of a mouse embryo fibroblast was obtained using indirect immunofluorescence techniques.313 The cells were fixed with formaldehyde, dehydrated, and treated with antibodies (formed in a rabbit) to microtubule protein. The cells were then treated with fluorescent goat antibodies to rabbit /-globulins (see Chapter 31) and the photograph was taken by fluorescent light emission. Courtesy of Klaus Weber. Interferon induction in normal and leukemic lymphocyte cultures with tilorone has been observed50. The interferon response observed in normal lymphocyte cultures appeared to be correlated with the toxic effect of tilorone. The effect observed in leukemic cultures required higher concentrations of tilorone, but, similarly, appeared to be related to cell viability. Tilorone has been reported to stimulate production of interferon by itself in mouse embryo fibroblasts and, in combination with poly rl poly rC/DEAE-dextran in mouse L929 cells51. Human foreskin fibroblasts were not stimulated. The degree of synergism between tilorone and the nucleotide-dextran complex was proportional to the concentrations of tilorone and poly rl/poly rC and was influenced by the times of addition of the compounds relative to each other. [Pg.131]

Enzyme activity was assayed in the high speed supernate obtained from rat livers homogenized in 50 mM potassium phosphate buffer (pH 7.4) containing 0.1 M NaCI. It was also shown to be applicable to extracts from human promyelocytic leukemia HL-60 cells, and mouse embryo fibroblasts. [Pg.269]

There have been some attempts to use spin labels to probe for differences in the properties of membranes of normal and tumour cells. Thus mouse embryo fibroblasts have been labelled with C (n = 4, m = 10) and it was found that on transformation with oncogenic DNA or RNA... [Pg.239]

There is a sizable database of studies investigating the DNA-damaging activity of mercuric chloride. The finding that mercuric chloride can damage DNA in rat and mouse embryo fibroblasts (Zasukhina et al. 1983), supports the in vivo evidence of species- and intraspecies-specific sensitivity to the genotoxic action of mercuric chloride. Marked conversion of DNA into the single-stranded form occurred at 10 6 M... [Pg.318]

Nocodazole, a drug that inhibits the assembly of microtubules, arrests normal mouse embryo fibroblasts with a 4N content, whereas fibroblasts from p53-deficient mouse embryos undergo several rounds of DNA synthesis without chromosome segregation (Cross et al., 1995). This spindle checkpoint differs from the G2/M checkpoint. Both p53+/+ and p53 / fibroblasts treated with nocodazole transiently arrest at mitosis for the same length of... [Pg.96]
