Responses in many of these assays were weak and occurred at toxic doses, and were affected by tissue culture conditions modifying uptake by the cell. The mechanism of nickel carcinogenesis is controversial, and is likely to vary with the form of nickel. The nickel ion (Ni + ) alone does not form premutagenic DNA lesions, suggesting that nickel causes indirect DNA damage, perhaps due to oxidative stress or blocking DNA repair mechanisms.

Injection site tumors are induced by many nickel compounds that do not cause cancer in animals by other routes of exposure. In fact, most of tihe published literature on nickel carcinogenesis concerns injected or implanted metallic nickel or nickel compounds these data are of limited value in determining carcinogenic exposure levels for avian and terrestrial wildlife.
