Aspart insulin

Human insulin is derived from a biosynthetic process using strains of Escherichia coli (recombinant DNA, rDNA). Human insulin appears to cause fewer allergic reactions than does insulin obtained from animal sources. Insulin analogy, insulin lispro, and insulin aspart are newer forms of human insulin made by using recombinant DNA technology and are structurally similar to human insulin. 

Insulin aspart is given immediately before a meal (widiin 5 to 10 minutes of beginning a meal). Insulin lispro is given 15 minutes before a meal or immediately after a meal. Insulin aspart and lispro make insulin administration more convenient for many patients who find taking a drug 30 to 60 minutes before meals bothersome hi addition, insulin lispro (Humalog) appears to lower die blood sugar level 1 to 2 hours after meals... 

A number of combination insulin products are available commercially. NPH is available in combinations of 70/30 and 50/50 with regular insulin. Two short-acting insulin analog mixtures are also available. Humalog Mix 75/25 contains 75% insulin lispro protamine suspension and 25% insulin lispro. Novolog Mix 70/30 contains 70% insulin aspart protamine suspension and 30% insulin aspart. The lispro and aspart insulin protamine suspensions were developed specifically for these mixture products and will not be commercially available separately.

When compared with soluble insulin, the human analogue insulin aspart has a faster onset and a shorter duration of action, resulting in a higher fasting and preprandial blood glucose concentration. The incidence of hypoglycaemia tends to be lower with insulin aspart than with soluble insulin.

Contains 70% insulin aspart protamine suspension and 30% insulin aspart (rDNA). 

Insulin aspart Because insulin aspart has a more rapid onset and a shorter duration of action than human regular insulin, insulin aspart normally should be used in regimens together with an intermediate or long-acting insulin. NovoLog may be infused subcutaneously by external insulin pumps. 

Because of the fast onset of action of insulin aspart, administer close to a meal (start of meal within 5 to 10 minutes after injection). 

Mixing of insuiins-The effects of mixing insulin aspart or lispro with insulins of animal source or insulin preparations produced by other manufacturers have not been studied (see Warnings). 

Insulin aspart If insulin aspart is mixed with NPH human insulin, draw insulin aspart into the syringe first. Do not mix insulin aspart with crystalline zinc insulin preparations. When used in external subcutaneous infusion pumps for insulin, do not mix with any other insulins or diluent. 

Insulin aspart, insulin glulisine - Localized reactions and generalized myalgias have been reported with the use of cresol as an injectable excipient. 

Lactation Lactating women may require adjustments in insulin dose and diet. It is unknown whether insulin glargine, insulin aspart, or insulin glulisine are excreted in significant amounts in breast milk.

Insulin Aspart is a second fast-acting engineered human insulin analogue now approved for general medical use. It differs from native human insulin in that the proline residue has been replaced by aspartic acid. This single amino acid substitution also decreases the propensity of individual molecules to self-associate, ensuring that they begin to enter the bloodstream from the site of injection immediately upon administration.

Insulin lispro, insulin aspart, insulin detemir, insulin glargine and insuline glulisine are human in-sullin analogues with the same mechanism of action. It should be noted however that the potency of insulin detemir was decreased four fold compared to human insulin. By changing amino acids on some locations absorption rates and the duration of action may be changed compared to human insulin.
