HMSCs

Burger, R., Schorn, C., and Bigler, P., HMSC simultaneously detected heteronuclear shift correlation through multiple and single bonds, J. Magn. Reson., 148, 88, 2001. [Pg.122]

Another study looked at the relationship between selected markers and hMSC-mediated immunosuppression in vitro. It was found that co-culture of hMSC with anti-CD3/CD28 activated peripheral blood mononuclear cells (hPBMCs) caused inhibition of lymphocyte proliferation. The effect of hMSCs on lymphocyte proliferation was dose dependent, causing more than 50% inhibition at an approximately 1 10 to 1 25 MSC lymphocyte ratio. Five-day... [Pg.63]

In October 2004, clinical data from a phase I safety trial of OTI-OIO therapy for the treatment of GVHD were reported. Infusion of hMSCs decreased GVHD in 22 to 45% of patients, suggesting that OTI-OIO reduces the numbers of pro-inflammatoiy cytokines and T-cells while increasing levels of anti-inflammatory cytokines in these patients [570810]. [Pg.65]

Stem cell isolation and expansion BM aspirates were combined with DPBS and centrifuged several times at different density gradients to collect hMSCs. Cells were then resuspended in hMSC complete culture medium and plated at 3 x 107 cells/185 cm2. The cultures were maintained at 37°C in a humidified atmosphere before being passaged prior to confluency. 643694"... [Pg.70]

In vitro Modulation of allogeneic immune cell responses by hMSC hMSCs incubated with DCs and naive T-cells When incubated with DC a > 50% decrease in TNFa secretion and a > 50% increase in IL-10 secretion were observed. hMSC decreased IFN. Secretion by Thl and NK cells and increased IL-4 secretion by Th2 cells when incubated with naive T-cells 643694"... [Pg.70]

In vitro Relationship between selected markers and hMSC-mediated immunosuppression Co-culture of hMSCs with anti CD3/CD28-activated hPBMCs A correlation was observed between PGE2 secretion and IDO enzyme activity on the inhibition of lymphocyte proliferation by hMSC 656696... [Pg.70]

In vivo HMSC engraftment and migration Fischer rat MSCs were infused intraportally into the livers of other Fischer rats (2 million cells/rat) Results showed cells to persist for at least 28 days, being localized to the portal triad regions 643859... [Pg.70]

In this study, the authors examined the immunomodulary functions of human MSCs by coculturing them with purified subpopulations of immune cells, and report that hMSCs alter the cytokine secretion profile of the host immune cells to induce a more antiinflammatory phenotype. The authors discuss and propose mechanisms underlying the immunomodulaiy functions of hMSCs in allogeneic transplantation. A shift from a proinflammatory environment toward an anti-inflammatory or tolerant cell environment would result in inflammation modulation, tolerance induction and reduction of allogeneic transplantation complications. [Pg.72]

Substrate topographical patterns have also been shown to affect the differentiation of MSCs to nerve cells. Yim et al. investigated the differentiation and proliferation of hMSCs on nanogratings of 350 nm width and formd alignment and elongation of... [Pg.46]

Li et al. [83] fabricated silk fibroin fiber scaffolds containing bone morphogenetic protein 2 (BMP-2) and/or nanoparticles of hydroxyapatite (nHAp) by electrospinning. These scaffolds were used in vitro to study bone formation from hMSCs. The results showed that the incorporation of BMP-2 and/or nHAp into silk... [Pg.222]

Instead of the destructive co-addition of unwanted coherence, in the HMSC experiment [5.207(b)], Check it 5.8.1.4, data relating to one-bond correlations and multiple-bond correlations are interleaved in a single data file. A pre-processing script... [Pg.342]

Dang et al. [167] developed a thermosensitive hydroxybutyl chitosan (HBC) and evaluated its compatibility with hMSCs. The HBC can well support hMSCs in terms of metabolic activity and ECM production. Another thermosensitive... [Pg.107]

HMSC - Heteronuclear Multiple-bond and Single-bond Coupling connectivities... [Pg.84]

Bigler and co-workers illustrated the application of the HMSC experiment using strychnine (14) as a model compound. Excellent discrimination in the processed data was obtained between the direct and long-range correlation responses. Results obtained with the HMSC experiment were compared to conventional HMBC and statically-optimized ACCORD-HMBC data. The sensitivity and lineshape were comparable to the HMBC data and superior to the statically-optimized ACCORD-HMBC data due to the evolution of antiphase coherence into in-phase coherence prior to detection, resulting in FlDs that are not optimal for magnitude calculated presentation. This observation is analogous to that made with the BIRD-HMBC data above (see Section 3.3.6). [Pg.85]
