After 5 years in the FDA s fast track development program, Romidepsin (Fig. 18) was approved by the FDA for refractory cutaneous T-cell lymphoma on November 6,2009. In the literature, romidepsin has also been called depsipeptide, FK228, FR901228, and NSC-630176. It was isolated from bacterial fermentation extracts from Chromobacterium violaceum and is a potent inhibitor of HDAC. In some human cancer cell lines, romidepsin inhibits HDACs at levels ten times that of TSA. [Pg.290]

Collections of synthetic compounds are easily screened in recombinant microbes [6,13], Fermentation extracts present more problems, since they may contain nutrients that reduce dependence on the target protein. It proved difficult to screen fermentation extracts in the ODC assay [40], since even trace amounts of polyamines in the extract eliminate dependence on the parasite enzyme. Conversely, the PFK assay was compatible with fermentations. The suitability of fermentation extracts must be empirically determined in each case. [Pg.334]

Cell recovery is mainly concerned with what is known as clear mash, i.e. sugar solutions without any entrained solids suchas as those in a whole-grain mash. The use of cell recovery on mash can be accomplished by clarifying the mash prior to fermentation or else incorporating a wet-milling front end and taking the saccharified starch as substrate. However the production of a clear mash directly from cereal mash is presently not feasible. This is due to excessive losses of carbohydrate in the cake or non-fermentable extract ranging from 10 to 20 per... [Pg.56]

Concentrated steep liquor is officially identified for feeding purposes as Condensed corn fermentation extractives. This product carries 25% protein content at 48% moisture. Its only feeding use is as an ingredient in liquid feeds for cattle, but it is also an excellent nitrogen source in nutrient media for antibiotic production.244... [Pg.426]

Information concerning the method of manufacture may be presented in the form of a flow chart. Supply a brief description of the methods of isolation (e.g., synthetic process, fermentation, extraction, recombinant deoxyribonucleic acid [DNA] procedure) and purification (solvent recrystallization, column chromatography, distillation). Include all synthetic pathways that have been adequately characterized during the investigational stages of drug development. [Pg.192]

The remaining water is finely distributed across the fat phase by a kneading or extrusion process. During this phase aroma compormds, for example, lactic acid fermentation extracts, can be added to modify the taste profile in case sweet cream was used to produce the butter. [Pg.447]

Libraries of synthetic combinatorial chemicals complement libraries of natural product metabolites, but do not duplicate or replace them. Examples of side-by-side screening of synthetic combinatorial chemicals and microbial fermentation extracts indicate that both sets of chemical diversity can provide unique leads. The quantity of leads generated by each method is irrelevant, if the quality of the lead is not considered. Therefore, scientific research should be focussed not on which type of library is better, but rather how to take advantage of both resources in a cost-effective and timely manner. [Pg.66]

The Streptomyces WEC 85E assay has been used to test fermentation extracts from two thousand actinomycete isolates. Over one himdred strains, representing possibly twenty-seven different species, were foimd to produce metabolites that affect the onset of sporulation. Purification and characterization of the active metabolites present in some of these extracts is currently in progress. [Pg.96]

The production of CPS by bacteria fermentation-extraction-purification procedures presently reaches a very efficient level. Purified CPS or LPS should be chemically modified for conjugation to a protein by a very complex procedure and give a final product that needs to be as defined as possible. If the production or purification of the carbohydrate material from the natural source is difficult and one bears in mind the complexity of producing a consistent glycoconjugate from a natural pol)mer then opportunities for modern synthetic carbohydrate chemistry are at hand. [Pg.2716]

The bottom fermentation is divided into main and secondary fermentation. For the main fermentation, the cooled-down flavouring, which contains approx. 6.5-18% malt extract, is combined in fermentation containers with yeast (approx. 1 litre per hectolitre) and fermented at a temperature of 6-10°C for 6-10 days within this period approx. 90% of the fermentable extract is transformed. [Pg.509]
