Ergotamine tartrate

The lungs large, permeable surface makes systemic deUvery possible. For example, an inhaler deUvers 360 )Tg per dose of aerosolized ergotamine tartrate [379-79-3] for migraine (54), and inhalant systems deUver anesthetic gases. Research is under way on the systemic deUvery of proteins and peptides through the lungs (57,58).

Lisuride dihydrogen maleate (h/Jj 30-35), methysergide maleate (h/ij 40-45), dihydroergotamine mesylate (hRf 45-50) and ergotamine tartrate (hRj 70-75) appeared as blue violet chromatogram zones on a yellow background. The detection limits — calculated for free base — were 3-4 ng substance per chromatogram zone.

Their reaction was tested on the individual components of the test mixture indole, ergotamine tartrate, ergotaminine and ergobasine [ergometrine) [3]. The results obtained were as follows ...

DHE does not appear to cause rebound headache, but dosage restrictions for ergotamine tartrate should be strictly observed to prevent this complication.

Ergotamine tartrate (lOg) is added to a stirred de aerated (nitrogen stream) solution of 38 g potassium hydroxide in 100 ml of methanol and 200 ml of water. The solution turns pink to red. The solution is heated to reflux and the methanol is slowly removed using a partial takeoff. Methanol is allowed to distill until the pot temperature reaches 90-95° C. The mixture is then maintained at total reflux until the evolution of ammonia ceases (hold pH paper in outlet of reflux condenser to test for ammonia). Nitrogen should be bubbled through the mixture to entrain the ammonia. 

Dissolve 175 g of KOH in 1,750 ml of water in a flask of 5 liters volume equipped with a reflux condenser and a gas inlet tube. If a stirring device is not required, it should be removed and the open neck stoppered. Heat the mixture to 80° under a stream of nitrogen and add 500 g of ergotamine tartrate. Hold the temp at 80° for 2Vi hours with bubbling from the nitrogen filled gas inlet tube. Pour the mixture into a 5 gallon polyethylene bucket (made from the same... 

Ergotamine tartrate Nausea and vomiting occur in up to 10% of patients. Numbness and tingling of fingers and toes muscle pain in the extremities pulselessness weakness in the legs precordial pain transient tachycardia or bradycardia localized edema itching. 

The effectiveness of dihydroergotamine (DHE) and ergotamine tartrate was demonstrated in old clinical trials which probably would not meet currently accepted criteria for good methodology. For ergotamine tartrate, the dose usually recommended is 1-2 mg/d not to exceed 6 mg/d or 10 mg/week. DHE, which has a low bioavailability, is only effective after parenteral administration (subcutaneous, intramuscular, intravenous, or nasal-spray administration). The recommended dose is 1-2 mg/d. 

Caffeine is often used in combination with ergotamine tartrate to improve digestive tract absorption. However caffeine has an antimigraine action itself.
