Naproxen sodium

Quite a few NSAlDs are available as OTC products. OTCformulations such as ibuprofen (Advil, Motrin, and Nuprin), ketoprofen (Orudis), and naproxen sodium (Aleve) are available to any consumer. The potential for misuse and abuse is high, especially when one is confronted by the large number of advertisements on television and in print heralding the wonderful benefits of these products. 

The phase transformation relationships for the solvatomorphs of naproxen sodium have been reported [71], The dihydrate phase is obtained upon crystallization from water, and a monohydrate phase could be prepared by the dehydration of the dihydrate phase in a desiccator (RH = 0%) for two days. The anhydrate phase could be obtained from either the monohydrate or dihydrate by drying the substance in an oven at 120 °C for two hours. Thermal analysis data was used to demonstrate the existence of two types of water in the dihydrate phase, and that each could be removed at a characteristic temperature.

Primary dysmenorrhea Diclofenac potassium, ibuprofen, ketoprofen, mefenamic acid, naproxen, naproxen sodium. 

Naproxen oniy (not naproxen sodium) - Total daily dose is approximately 10 mg/kg in 2 divided doses. 

Naproxen sodium - 825 mg, then 275 mg every 8 hours until attack subsides. 

Naproxen sodium - 550 mg, followed by 275 mg every 6 to 8 hours. Do not exceed a 1.375 g total daily dose. 

Eideriy Age appears to increase the possibility of adverse reactions to NSAIDs. The risk of serious ulcer disease is increased this risk appears to increase with dose. Ketorolac is cleared more slowly by the elderly use caution and reduce dosage. Pregnancy Category B (ketoprofen, naproxen, naproxen sodium, flurbiprofen, diclofenac, fenoprofen, ibuprofen, indomethacin, meclofenamate, sulindac). 

Dermatologic effects Promptly discontinue mefenamic acid if rash occurs. A combination of dermatologic and allergic signs and symptoms suggestive of serum sickness occasionally have occurred in conjunction with the use of piroxicam. Concomitant therapy Do not use naproxen sodium and naproxen concomitantly both drugs circulate as naproxen anion.

The choline salts of naproxen and tolmetin yielded marked solubility enhancements, even over that of the sodium salts. The aqueous solubility of the choline salt of naproxen was 6700 times the solubility of naproxen and almost twice that of naproxen sodium. The choline salt of tolmetin was almost 8000 times as soluble as the parent drug and almost 5 times as soluble as its sodium salt (Murti, 1993) (Cheenu dissertation, our unpublished results). Table 15.2 shows that the melting points and enthalpies of fusion are both affected by the formation of the choline salt, and in fact, the choline salt proved to be more thermally stable.

Naproxen sodium tablets were used in an in vivo-in vitro evaluation with four different polymeric dispersions, cellulose acetate phthalate (CAP), cellulose acetate trimellitate (CAT), 50 50 CAP/CAT, and methacrylic acid copolymer [64], The study indicated that coating material that dissolves at a more acidic pH in vitro (such as CAT at pH 4.5) will also dissolve at a more acidic pH in vivo (i.e., the coating dissolves higher up in the GI tract). In addition, it was found that aging did not markedly affect dissolution characteristics of CAT or methacrylic acid copolymer-coated tablets.
