Eosinophil chemotactic factor

Alveolar macrophages release a number of inflammatory mediators, including PAF and leukotrienes B4, C4, and D4. Production of neutrophil chemotactic factor and eosinophil chemotactic factor furthers the inflammatory process.

Initial stimulation of the mast cell by any of a variety of agents causes the release of preformed histamine (H) neutrophil and eosinophil chemotactic factors (NCF, ECF) and enzymes and the de novo synthesis of prostaglandins (PG) and leukotrienes (LT). These agents increase vascular permeability and vessel diameter. As a result, albumin and later neutrophils (PMN) enter the tissue space where the latter undergo phagocytosis and the secretion of proteolytic enzymes to the extracellular space where they act on albumin to generate NRP (neurotensin-related peptide) and HRP (histamine-releasing peptide). These newly formed peptides then act as a second stimulus to the mast cell. In addition NRP and HRP may affect other immunocompetent celt such as monocytes, macrophages or eosinophils.

Eosinophils are attracted by proteins released by T cells, mast cells and basophils [eosinophil chemotactic factor of anaphylaxis (ECF-A)]. They bind schistosomulae coated with IgG or IgE, degranulate and release major basic protein, which is toxic. Eosinophils also release histaminase and aryl sulfatase, which inactivates histamine and Slow reacting substance of anaphylaxis (SRS-A). This results in antiinflammatory effects and inhibits migration of granulocytes to the site of injury.

With each blink the antigen-coated contact lens mechanically traumatizes the tarsal conjunctiva. This process causes the release of mediators, such as neutrophil chemotactic factor and eosinophil chemotactic factor, which attract inflammatory cells (e.g., neutrophils, eosinophils, mast cells, and basophils). The immunologic sequence of events results in an increase in tear immunoglobulins IgE and IgG and C3 anaphylatoxin. The tear immunoglobulins and C3 anaphylatoxin then interact with the inflammatory cells produced from the mechanical trauma. This interaction causes the release of vasoactive amines, resulting in subsequent clinical manifestations. Papillae formation is related to structural changes in the conjunctival epithelium and stroma associated with increased eosinophils and inflammatory cells.

Concerning mediators of immediate hypersensitivity, the most important include histamine, leukotriene e.g., SRS-A (slow-reacting substance of anaphylaxis, ECF-A (eosinophil chemotactic factor of anaphylaxis), PAF (platelet-activating factor), and serotonin. There are also three classes of lipid mediators that are synthesized by activated mast cells through reactions initiated by the actions of phospholipase A2. These are prostaglandins, leukotrienes, and platelet-activating factors (PAF). 

The eosinophil chemotactic factor of anaphylaxis (ECP-A) is a preformed product pund in rat mast cell granules, in human lung, nasp polyps, and PMN. The activity resides in two acidic tetrapeptides, Ala-Glp Ser-Glu and Val-Gly-Ser-Glu, which preferentially attract eosinophils. Synthetic peptides of this gomposition are equipotent to natural ECF-A botjj in vitro and in vivo. ECF-A shopg maximal chemotactic activity at 10 M, and concentrations as low as 10 M deactivate eosinophils tp sph-sequent stimulation by chemotactic concentrations of ECF-A and C5a. ...
