When attempting to determine minor impurities in a chemical process the concept of dynamic range of the detection technique is a more useful concept than sensitivity, because sensitivity is usually limited by chemical interference. It remains the case that the dynamic range of the HPLC/UV combination is the highest of all the commonly available analytical techniques, and therefore when it is essential to determine minor impurities online then this is still the preferred combination, despite its interfacing difficulties. The use of online HPLC is further discussed in the chromatography section. [Pg.252]

An example of how ES-MS can be used to determine minor impurities in a recombinant protein is shown in Figure 9.33, where some small additional ions in the mass spectrum of recombinant insulin-like growth factor (IGF) can be seen. Calibration and quantification procedures are easier in LA-ICP-MS compared to other solid-state mass spectrometric techniques because the laser ablation and the ICP ion source operate at normal pressure and the laser ablation of solid samples and ionization of analytes are separated in space and time. Therefore the advantage of solution calibration in ICP-MS can be applied in this solid-state analytical technique. The introduction of solution based calibration, which is only possible in LA-ICP-MS, was an innovative step in the development of this sensitive mass spectrometric technique. A difference measurement is a technique that may be used to determine minor constituents. The major component is introduced into the comparison cell (a variable space cell may be useful here) at a concentration equal to its concentration in the sample cell. Its absorption is thereby compensated and the result is a spectrum of the minor constituents. There are several difficulties that must be appreciated when applying this technique. Because of strong absorption in both beams a wider than normal slit width must be used and this will result in poorer resolution and a deviation from Beer s Law. Care must be taken to avoid over- or under-compensation, not only for the major component, but also for the solvent if working in solution.
